摘　　要：目的探讨lnc RNA XLOC006926调控肝癌细胞增殖和转移机制,结合肝癌患者临床病理特征,分析其临床意义。方法利用实时荧光定量PCR方法检测lnc RNA XLOC006926在肝癌细胞系及肝癌患者配对(癌、癌旁)组织(n=88)中的表达水平。进一步通过CCK-8、克隆形成、划痕及transwell实验检测过表达或抑制lnc RNA XLOC006926对细胞增殖、迁移和侵袭的影响,同时结合临床信息统计分析其与临床病理特征及患者预后的相关性。结果 lnc RNA XLOC006926在肝癌细胞及肝癌组织中的表达显著降低,过表达lnc RNA XLOC006926后肝癌细胞增殖、迁移和侵袭能力降低,相反抑制lnc RNA XLOC006926后肝癌细胞增殖、迁移和侵袭能力增强。结合患者临床信息分析,lnc RNA XLOC006926的表达量与肿瘤大小(P=0.007)、PVTT(P=0.024)具有显著相关性,其低表达预示着预后较差。结论 lnc RNA XLOC006926在肝癌发生发展中发挥了重要作用,参与调节肝癌细胞的增殖及迁移,有望成为肝癌发生发展的生物标志物和肝癌预防治疗的新靶点。To investigate the mechanisms of lnc RNA XLOC006926 regulating the proliferation and metastasis of hepatocellular carcinoma（HCC） cells and to analyze its clinical significance, we determined lnc RNA XLOC006926 expression levels in paired HCC tissues（cancer, paired non-cancer）（n=88） and HCC cell lines by real-time fluorescence quantitative PCR. Effect of lnc RNA XLOC006926 on the proliferation, migration and invasion of HCC cells was estimated by CCK-8, wound healing, and Transwell assays. And the association between lnc RNA XLOC006926 and the overall survival of patients was analyzed by statistical analysis. Data showed that the expression of lnc RNA XLOC006926 in HCC tissues was significantly decreased, while overexpression of lnc RNA XLOC006926 could inhibit the proliferation, migration and invasion of HCC cells. On the contrary, knockdown of lnc RNA XLOC006926 enhanced the ability of proliferation, migration and invasion of HCC cells.According to the clinical information of patients, the expression level of lnc RNA XLOC006926 was significantly correlated with the tumor size（P=0.007）and PVTT（P=0.024）, and the lower expression of lnc RNA XLOC006926 indicated poor prognosis. Collectively, our findings suggest that lnc RNA XLOC006926 plays an important role in the development of HCC by participating in carcinoma cell proliferation and migration. Thus lnc RNA XLOC006926 might be a powerful candidate prognostic biomarker and therapeutic target of HCC.
