摘　　要：目的:探究卷烟烟气对他莫西芬在大鼠乳腺癌中抗癌作用的影响。方法:利用N-甲基-N-亚硝基脲单次腹腔注射建立大鼠乳腺癌模型,设定他莫西芬单次给药和多次给药两个大组,大组中分别设立空气组、烟气组和空白溶剂对照组。通过ELISA试验和免疫组化方法分别测定大鼠血清和肿瘤组织中8-羟基脱氧鸟苷(8-OHd G)的含量,同时用ELISA测定肿瘤组织中抗细胞核增殖抗原(PCNA)的含量。结果:成功诱导构建了大鼠乳腺癌模型;他莫西芬能显著降低血清和肿瘤组织中8-羟基脱氧鸟苷的含量并抑制肿瘤组织中PCNA的表达;而合并烟气给药会相对升高8-羟基脱氧鸟苷和PCNA的表达。结论:烟气会减弱他莫西芬在大鼠乳腺癌中的抗增殖能力。Objective： To explore if the cigarette smoke may influence the anti-tumor effect of tamoxifen. Methods： The breast cancer rat models were established by single intraperitoneal injection of N-methyl-N-nitrosoure.All the model rats were divided into two groups： Single dose and multiple dose of tamoxifen groups. And every group consisted of tamoxifen ＋ air,tamoxifen ＋ cigarette smoke and solvent ＋ air sub-groups. Detect the expression of 8-hydroxy-2-deoxyguanosine（ 8-OHdG） in the rats serum and tumors by ELISA and immunohistochemical analysis respectively. And explore the expression of proliferating cell nuclear antigen（ PCNA） in the breast cancer tissues using ELISA. Results： The rat breast cancer models were established successfully. And tamoxifen could down-regulated the expression of 8-OHdG in both rats serum and mammary tumor tissues. In the meanwhile,the expression of PCNA in the cancer tissues were also inhibited after administration of tamoxifen. However,when combined with cigarette smoke,the expression of 8-OHdG and PCNA were increased. Conclusion： Cigarette smoke could weaken the inhibition effect of tamoxifen on the cell proliferation in breast cancer rats.
