房角关闭对兔眼小梁组织MMP-2、TIMP-2及AQP-1表达的影响

摘　　要：目的建立兔眼房角关闭模型,比较基质金属蛋白酶-2(MMP-2)、金属蛋白酶组织抑制因子-2(TIMP-2)、水通道蛋白-1(AQP-1)在对照组和模型组表达水平的变化,探讨房角关闭对小梁网功能变化的影响。方法实验研究。健康成年新西兰大白兔24只(48只眼)随机分为4组,每组6只(12只眼),每只随机选取一眼作为实验组,予玻璃体腔注入透明质酸钠联合前房放液,建立房角关闭模型,另一眼不作处理为对照组,应用免疫荧光和计算机图像分析技术分别于造模成功后1个周、1个月、2个月、4个月观察各组MMP-2、TIMP-2、AQP-1的表达情况。结果正常对照组和各实验组均有MMP-2、TIMP-2、AQP-1的表达。MMP-2在实验组的表达量随造模时间延长先升高后降低,各组与正常对照组相比均有明显差异(P<0.05);TIMP-2在实验组的表达量随造模时间延长先升高后降低,各组与正常对照组相比均有明显差异(P<0.05);MMP-2/TIMP-2比值在造模后1个周、1个月组与正常对照组相比无明显差异(P>0.05),而2个月组、4个月组与正常对照组相比差异明显(P<0.05);AQP-1在实验组的表达量随造模时间延长先升高后降低,各组与正常对照组相比均有明显差异(P<0.05)。结论房角关闭早期,小梁网因代偿机制,仍存有部分功能,应尽早采取干预措施。Objective To establish a rabbit model of angle-closure and to investigate the expression of MMP-2、TIMP-2、AQP-1 in trabecular meshwork. Methods 24 New Zealand albino rabbits were randomly and equally divided into four groups. One eye of each rabbit was randomly selected to receive intravitreous injection of sodium hyaluronate and anterior chamber penetration. The fellow eye received no intervention. Expression of MMP-2,TIMP-2,AQP-1 were analyzed by immunofluorescence techniques and computer image analysis at 1 week,1 month,2 and 4 months. Results MMP-2,TIMP-2 and AQP-1 were detected in both treated eye and fellow eye. MMP-2,TIMP-2 and AQP-1 increased and then decreased following the treatment when compared to the fellow eyes（ all P 0. 05）. MMP-2/TIMP-2 ratio was similar to that of the fellow eyes at 1 week and 1 month but was significantly different at 2 or 4 months（ P 0. 05）. Conclusions Trabecular meshwork function is compensated up to 1 month after angle-closure. This is the time window when appropriate intervention should be done.
