恩度联合顺铂对非小细胞肺癌细胞FOXP3表达和凋亡的影响

摘　　要：目的探讨恩度联合顺铂对非小细胞肺癌(NSCLC)细胞叉头/翅膀状螺旋转录因子(FOXP3)表达和凋亡的影响。方法培养NSCLC细胞A549,将细胞分均为四组,分别加入恩度200μg/ml(恩度组)、顺铂2μg/ml(顺铂组)、恩度200μg/ml和顺铂2μg/ml(联合组)以及PBS 10μl(对照组)。48h后,检测FOXP3和Caspase-3的表达和细胞凋亡情况。结果处理A549细胞48h后,恩度组及联合组FOXP3蛋白表达量较对照组减少(P<0.05),而顺铂组FOXP3蛋白表达较其他组增加(P<0.05)。恩度组、顺铂组和联合组细胞凋亡和Caspase-3、剪切型Caspase-3的表达较对照组高,且联合组最高(P<0.05)。结论恩度联合顺铂能减少FOXP3的表达,促进NSCLC细胞凋亡,从而杀伤肿瘤细胞。Objective To investigate the effects of endostar combined with cisplatin on the expression of forkhead/winged helix transcription factor（FOXP3） and the apoptosis of non-small cell lung cancer （NSCLC） cells. Methods The NSCLC cell A549 was cultured and then treated with endostar 200μg/ml（group A）, cisplatin 2μg/ml（group B）, endostar combined with cisplatin（group C） and PBS 10 /11 （group D）. The expressions of FOXP3 and Caspase-3 and cell apoptosis were analyzed 48 hours later. Results The protein expression of FOXP3 was lower in groups of A and C than that in group D, which was higher in group B than other groups（P〈0. 05）. The cell apoptosis and expressions of Caspase-3 and cleaved Caspase-3 were higher in groups of A, B and C than those in group D,which were the highest in group C（P〈0. 05）. Conclusion Endostar combined with cisplatin can decrease the expression of FOXP3 and promote the cell apoptosis, which could kill the tumour cells.
