摘　　要：目的探讨微小RNA-1271(miR-1271)对胰腺癌中Rab34表达的影响。方法收集22例胰腺癌患者癌组织和对应癌旁组织标本。采用qRT-PCR检测癌组织和癌旁组织Rab34和miR-1271mRNA表达水平,IHC和Western blot检测Rab34蛋白表达水平。细胞转染法检测miR-1271模拟物对胰腺癌细胞Rab34表达的影响,双荧光素酶报告基因检测系统验证miR-1271结合Rab34的3′非翻译区靶位点。结果与癌旁组织相比,Rab34mRNA和蛋白在胰腺癌组织中表达升高(P<0.05),miR-1271mRNA表达降低(P<0.05)。miR-1271模拟物转染后抑制了胰腺癌细胞Rab34蛋白的表达。Rab34是miR-1271作用的靶基因。结论 Rab34可能是胰腺癌的分子标志物,miR-1271可调控其表达。Objective To investigate the effect of microRNA-1271（miR-1271） on expression of Rab34 in pancreatic cancer. Methods The cancer tissues and tumor-adjacent normal tissues from 22 pancreatic cancer patients were collected for detecting the mRNA expressions of Rab34 and miRq271 by qRT-PCR and Rab34 protein expression by immunohistochemistry and Western blot, respectively. Cell transfection was used to discover the regulation of miR-1271 on Rab34 gene. Double luciferase reporter assay was performed to identify miR-1271 binding to the 3＇ untranslated region of the Rab34 gene. Results Compared with tumor-adjacent normal tissues, the mRNA and protein expressions of Rab34 were increased and miR-1271 mRNA expression was decreased in pancreatic cancer tissues （P〈0. 05）. The expression of Rab34 protein was inhibited in pancreatic cancer cells after transfected with miR-1271 mimics. Rab34 was identified as the target gene of miR-1271. Conclusion Rab34 may be a tumor biomarker of pancreatic cancer, miR-1271 is able to regulate Rab34 expression.
