摘 要:目的探究奥卡西平(OXC)在戊四氮诱导癫痫小鼠模型治疗中对Caspase-3蛋白表达和细胞凋亡的影响作用。方法 60只雌雄各半的昆明小鼠,随机分为3组,每组各20只,雌雄各半。PTZ(癫痫)组:PTZ致癫痫+0.9%生理盐水灌胃;OXC(给药)组:PTZ致癫痫+OXC灌胃;CON(对照组):0.9%生理盐水+0.9%生理盐水灌胃组。在给药0.5 d、3 d、7 d、12 d每组分别处死5只小鼠,取脑组织进行免疫组化分析并全程记录小鼠抽搐癫痫情况。结果和CON组比较,PTZ和OXC组均出现显著的抽搐和癫痫症状(P<0.05),PTZ和OXC两组比较,PTZ组出现较高强度的(≥Ⅲ级)的抽搐症状,特别在Ⅴ-Ⅵ级中,OXC抽搐程度显著低于PTZ组(P<0.05)。在给药后的0.5 d、3 d、7 d和12 d不同时间点中,PTZ和OXC组的Caspase-3细胞光密度显著高于CON组(P<0.05),组间比较,在0.5 d和3 d中,PZT和OXC组的Caspase-3差异无统计学意义(P>0.05),在7 d和12 d中,OXC组的Caspase-3细胞光密度则显著低于PTZ组(P<0.05)。i NOS细胞光密度比较显示,OXC在给药后3 d后,i NOS细胞光密度值显著低于PTZ组(P<0.05)。PTZ和OXC组的Bax和Bcl-2阳性细胞数量均显著高于CON组(P<0.05),但是两组比较显示,给药3 d后,OXC组的Bax阳性细胞数量显著低于PTZ组(P<0.05),在给药后7d,OXC组的Bc1-2阳性细胞数量显著低于PTZ组(P<0.05)。结论奥卡西平对戊四氮诱导小鼠癫痫模型中的神经损伤具有保护作用,其可能是通过降低小鼠机体Bax家族相关蛋白表达进而降低了Caspase-3蛋白的表达。Objective To explore the effect of oxcarbazepine (OXC) inducing effect on apoptosis and expression of Caspase-3 protein in the mouse model of epilepsy in n e four. Methods 60 male and female half of Kunming mice were ran- domly divided into 3 groups, 20 rats in each group, half and half of male and female. PTZ (epilepsy) group : PTZ epilepsy, +0.9% physiological saline gavage; OXC (drug) group: PTZ epileptic +OXC garage; CON (control group) : 0.9% saline + 0.9% physiological saline gavage group. After administration of 0.5 d, 3D, 7 d and 12D, 5 mice were killed in each group. The brain tissues were analyzed by immunohistochemistry, and the seizures and seizures of the mice were recorded. Re- suits compared with group CON, PTZ and OXC group showed significant symptoms of convulsion and epilepsy (P〈0.05), PTZ and OXC between the two groups, high strength PTZ group (over grade III) tic symptoms, especially in the V - VI class, OXC tic degree was lower than that of group PTZ (P〈0.05). After the administration of 0.5 d, 3D, 7 d and 12D at different time points, Caspase-3 cell optical density of PTZ and OXC were significantly higher than that of CON group (P〈 0.05), comparison between groups, 0.5 d and 3D, PZT and OXC in group Caspase-3 had no statistical significance (P〉 0.05), 7 d and 12D, Caspase-3 the cell optical density of group OXC was significantly lower than that of PTZ group (P〈 0.05 ). Comparison of iNOS cell optical density showed that the light density of iNOS ceils after OXC administration was sig- nificantly lower than that in group PTZ (P〈0.05) after 3D administration. PTZ and OXC group the number of Bax and Bcl-2 positive cells were significantly higher than that of CON group (P〈0.05), but the two groups showed that after administra- tion of 3D OXC group, the number of Bax positive cells was significantly lower than that of PTZ group (P〈0.05), after ad- ministration of 7D, OXC group the number of Bcl-2 positive cells was si
