摘  要:目的:探讨miR-199a-3p调控卵巢癌细胞侵袭转移及机制。方法:QPCR技术检测22例卵巢癌组织及20例卵巢正常组织中DDR2 mRNA表达水平。采用miR-199a-3p mimics转染人卵巢癌SKOV3细胞,QPCR技术检测DDR2 mRNA表达,Western blot法检测DDR2、Wnt/β-catenin相关通路信号分子表达。结果:DDR2在卵巢癌组织中表达明显升高。较SKOV3细胞组,miR-199a-3p mimics组中DDR2 mRNA表达水平降低,DDR2、β-catenin、c-myc、c-JUN表达水平明显下调。结论:miR-199a-3p可能通过下调DDR和Wnt/β-catenin通路抑制卵巢癌细胞体外侵袭转移。Objective: To investigate the effects and mechanism of miR-199 a-3 p regulating the invasion and metastasis of ovarian cancer cells. Methods: The DDR2 mRNA expression level of 22 cases of ovarian cancer tissues and 20 cases of normal ovarian tissues were measured with QPCR.Human ovarian cancer cell line SKOV3 was transfected with miR-199 a-3 p mimics.QPCR was used to detect the DDR2 mRNA expression.Western blot was used to detect the protein expression levels of signal pathways of DDR2 and Wnt/β-catenin. Results:DDR2 expression was significantly upregulated in ovarian cancer tissue. DDR2 mRNA expression level was significantly decreased in miR-199 a-3 p mimics group,and the levels of DDR2,β-catenin,c-myc,and c-JUN protein were significantly decreased.Conclusions: miR-199 a-3 p may inhibit the invasion and metastasis of ovarian cancer cells by decreasing DDR2 and inhibiting Wnt/β-catenin pathway.
