摘 要:目的研究LASP-1是否影响肿瘤来源成骨样细胞和非肿瘤来源成骨细胞的运动、成骨、增殖能力以及细胞周期的分布比例。方法采用qRT-PCR及Western Blot(WB)检测小鼠骨髓间充质干细胞(BMMSC)、人成骨细胞(h OB)以及人骨肉瘤细胞(MG-63)中LASP-1的表达;通过构建重组载体,转染sh-LASP-1,检测各细胞中LASP-1表达的变化,采用划痕实验、XTT法、WB、流式细胞仪检测该变化对细胞运动能力、骨钙素(OCN)表达、细胞增殖能力和细胞周期分布比例的影响。结果上调或下调LASP-1的表达,非肿瘤来源细胞(BMMSC,h OB)的运动能力、骨钙素(OCN)的表达、细胞增殖能力及G2/M期的细胞比例未见明显改变;对肿瘤来源细胞(MG-63)而言,随LASP-1的表达上调,OCN表达亦上调,细胞数量增多,且处于G2/M期的细胞比例降低;对各细胞运动能力未见显著影响。结论 LASP-1对非肿瘤来源细胞的成骨性能、增殖能力以及细胞周期分布比例未见明显影响;而对肿瘤来源细胞(MG-63)有明显调节作用。Objective To investigate the impacts of LASP-1 on the behavior and cell proliferation abilityof tumor derived osteoblast-like cells and non tumor derived osteoblasts. Methods QRT-PCR and Western blotwere performed to detect LASP-1 expressions in mouse bone marrow mesenchymal stem cells(BMMSC),humanosteoblasts(h OB)and human osteosarcoma cell(MG-63). With the construction of recombinant vector and thetransfection of sh LASP-1,the expression changes in both LASP-1 and osteocalcin(OCN),and the cell number ofcell-cycle phase distribution were analyzed. Results With up-or down-regulation of LASP-1,osteocalcin(OCN)expression,cell proliferation ability and the cell number of G2/M phase in non tumor cells(BMMSC,h OB)hadno obvious changes. However,as for the tumor cells(MG-63),with an increased expression of LASP-1,the ex-pression of OCN,cell proliferation number and the cell number at G2/M phase changed. There was no significanteffect on the moving ability of each cell. Conclusions LASP-1 has no significant effect on the osteogenic proper-ties,cell proliferation ability and cell cycle distribution of non tumor derived cells,but has a regulatory effect ontumor derived cells(MG-63).
