摘 要:目的探讨中药复方糖肾安对实验性2型糖尿病大鼠肾组织核因子NFE2相关因子(Nrf2)/抗氧化反应元件(ARE)通路的影响。方法将50只健康雄性SD大鼠随机分为正常组8只和造模组42只,造模组高脂高糖喂养,4周后按35 mg/kg一次性腹腔注射链脲佐菌素(STZ)建立2型糖尿病模型。将成模大鼠随机分为模型组、糖肾安低剂量组、糖肾安中剂量组、糖肾安高剂量组及西药组。各给药组给予相应的剂量药物灌胃,8周后收集所有大鼠肾脏标本。采用免疫组化技术检测大鼠肾组织中Nrf2、血红素加氧酶(HO-1)表达情况,Western blot、RT-PCR技术检测大鼠肾组织中核因子κBp65(NF-κBp65)表达情况,ELISA法检测大鼠肾组织中超氧化物歧化酶(SOD)活性。结果模型组大鼠肾组织中Nrf2、HO-1、NF-κBp65表达水平均明显高于正常组(P均<0.05),而SOD活性明显弱于正常组(P<0.05);糖肾安中剂量组、糖肾安高剂量组及西药组大鼠肾组织中Nrf2、HO-1表达水平及SOD活性均明显高于模型组(P均<0.05),而各给药组大鼠肾组织中NF-κBp65表达水平均明显低于模型组(P均<0.05)。结论中药复方糖肾安可激活糖尿病大鼠肾脏Nrf2/ARE通路,抑制NF-κBp65活化,增强抗氧化酶活性,减轻糖尿病大鼠肾脏氧化应激及炎症反应,从而发挥肾脏保护作用。Objective It is to investigate the effect of Tangshen'an Recipe on the Nrf2/ARE pathway in renal tissue of experimental type 2 diabetic rats. Methods 50 healthy male SD rats were randomly divided into normal group( n = 8) and molding group( n = 42),the rats in the molding group were treated with high fat and high glucose for 4 weeks. Type 2 diabetes model was established by intraperitoneal injection of streptozotocin( STZ) at a dose of 35 mg/kg. The molding rats were randomly divided into model group,low,medium,high dose of Tangshen'an group and western medicine group.Each drug group was given corresponding treatment by gavage,and the kidney specimens were collected after 8 weeks. The expression of Nrf2 and HO-1 in rat kidney tissue was detected by immunohistochemistry. The expression of NF-κBp65 and the activity of SOD in rat kidney were detected by western blot and RT-PCR. Results Compared with the normal group,the expression of Nrf2,HO-1 and NF-κBp65 in the renal tissue of the model group was increased,while the activity of SOD was decreased significantly( P<0. 05). Compared with the model group,the expression of Nef2,HO-1 and SOD in medium and high dose of Tangshen'an groups and western medicine group was significantly increased,while the expression of NF-κBp65 in each treatment group was decreased( P<0. 05). Conclusion Chinese medicine Tangshen'an compound prescription can activate Nrf2/ARE pathway in diabetic neonatal kidney,inhibit NF-κBp65 activation,enhance antioxidant enzyme activity,thereby reducing the diabetic rats kidney inflammation and oxidative stress response,and thus play a role in protecting the kidneys.
