FAM96A在人肝癌组织中的表达及其与临床病理特征的关系

摘　　要：目的检测96序列相似的家庭成员A(FAM96A)在人肝癌组织中的表达及其与肝癌生物学特征的关系。方法采用实时荧光定量聚合酶链反应(Real-time PCR)及Western blot法检测FAM96A蛋白在肝癌组织和癌旁组织、Hep G2细胞和L02细胞中的mRNA及蛋白表达量,比较其差异,并分析FAM96A蛋白表达差异与肝癌临床病理特征之间的关系。结果 Real-time PCR检测结果显示,FAM96A在肝癌组织中的mRNA表达低于癌旁组织(t=21.773,P<0.001),在Hep G2细胞中的表达低于L02细胞(t=26.153,P<0.001)。同时,Western blot检测结果证实:与癌旁组织相比,FAM96A在肝癌组织中的蛋白表达显著降低(t=15.582,P<0.001),在Hep G2细胞中的表达同样低于L02细胞(t=14.478,P<0.001)。且FAM96A蛋白表达差异与本研究中48例肝癌患者肿瘤大小、是否合并门静脉癌栓、是否发生肝外转移、Child分级、巴塞罗那肝癌分期之间存在相关性(P<0.001),然而与患者的性别、年龄、肿瘤个数、是否合并乙肝、是否合并肝硬化、血清甲胎蛋白无明显相关(P>0.05)。结论 FAM96A在人肝癌组织中表达显著下降,其表达水平与肝癌的发生发展、侵袭转移密切相关。Objective To investigate the expression difference of family with sequence similarity 96,member A（ FAM96 A） in 48 cases of liver cancer and explore the relation between FAM96 A expression and clinicopathological characteristics of liver cancer. Methods Real-time PCR and Western blot were used to examine the expression and distribution of FAM96 A in liver cancer tissues and their adjacent tissues,Hep G2 cells and L02 cells. Furthermore,the correlation between the expression difference of FAM96 A and clinicopathological characteristics of liver cancer was analyzed. Results FAM96 A mRNA levels was significantly decreased in liver tissues than in adjacent tissues（ t = 21. 773,P 0. 001） and in Hep G2 cells and L02 cells（ t = 26. 153,P〈0. 001）,while FAM96 A protein expression levels was lower in liver tissues compared with adjacent tissues（ t = 15. 582,P〈0. 001） and lower in Hep G2 cells and in L02 cells（ t = 14. 478,P〈0. 001）. Moreover,the expression difference was correlated with tumor size,portal vein tumor thrombosis,Child-Turcotte-Pugh,extrahepatic metastasis,Barcelona Clinic Liver Cancer（ BCLC） classification（ P〈0. 001）,but not with gender,age,tumor number,hepatitis B virus,cirrhosis of the liver and serum AFP of patients. Conclusion The FAM96 A protein expression is significantly decreased in48 cases of liver cancer tissues than in adjacent normal tissues. In addition,FAM96 A may be a good biomarker to evaluate the development,invasion and metastasis of liver cancer.
