摘 要:目的研究不同病理类型肺癌组织中Let-7的表达变化,重点探讨Let-7a在肺癌中的调控作用。方法收集腺癌、鳞癌、小细胞肺癌及癌周正常组织标本,用实时荧光定量PCR检测不同组织中Let-7的8个成员及不同病理分级下Let-7 a的表达情况。结果 (1)在同一肺癌组织或者正常组织标本中,Let-7的8个成员表达水平相近,结果差异无统计学意义(P>0.05);但与正常组织相比,癌组织中Let-7的8个成员的表达水平显著降低,差异有统计学意义(P<0.05)。(2)肿瘤组织中Let-7 a的表达水平在腺癌、鳞癌和小细胞肺癌中均低于正常组织。(3)在腺癌、鳞癌和小细胞肺癌组织中,肿瘤恶化程度越高,Let-7a表达越低;且Ⅰ级恶化、Ⅱ级恶化、Ⅲ级恶化之间差异有统计学意义(P<0.05)。结论 Let-7家族的8个成员均与肺癌发生、发展有密切关系,Let-7a的表达量可以指示肺癌恶化程度。Objective To investigate the expression of Let -7 in different pathological types of lung cancer tis- sues focusing on the effect of Let - 7a in lung cancer. Methods The specimens of adenocarcinoma, squamous carcino- ma, small cell lung cancer and adjacent normal tissues were collected. Real - time fluorescence quantitative PCR was used to detect the 8 members of Let -7 in different lung cancer tissues and Let -7a expression in specimens of different patho- logical grades. Results Firstly, in the same lung cancer tissue or normal tissue, expressions of eight members were simi- lar (P 〉0. 05 ). However, the expressions of 8 members in normal tissues were significant higher than that in the cancer tissue. Secondly, in adenocarcinoma, squamous carcinoma and small cell lung cancer, the expression of Let - was lower than that in normal tissues. Thirdly, in adenocarcinoma, squamous carcinoma and small cell lung cancer, expression of Let -7a was decreased with increased pathological grades, and there was a significant difference among grade Ⅰ , Ⅱ, Ⅲ deterioration. Conclusion There is a close correlation between all 8 members of Let -7 and lung cancer, which indicates that the expression of Let- 7a could reflect the pathological grades of lung cancer.
