摘 要:目的研究短波紫外线病毒灭活技术处理血小板后其体外功能相关参数的变化。方法白膜法提取血小板,将血小板重悬于PPP、SSP+混合溶液中,调血小板计数约1 000×109/L。短波紫外线照射处理血小板,辐照剂量为0.220 J/cm2。照射处理后扫描电镜观察血小板形态的变化,d1、d3、d5、d7取样检测血小板体外功能相关指标,评估短波紫外线病毒灭活技术对血小板体外功能的影响。结果扫描电镜观察到短波紫外线照射后血小板外形极不规则,伪足伸出,有激活的趋势。实验组血小板相较于对照组计数下降、MPV增加、P-LCR增加,HSR下降,差异均有统计学意义(P〈0.05);CCK-8法观察血小板线粒体脱氢酶的活性,实验组吸光度值稍低于对照组,d5差异有统计学意义(P〈0.05);实验组血小板激活标志PAC-1、CD62p,凋亡标志Annexin V阳性表达率均高于对照组,差异均有统计学意义(P〈0.05);实验组血小板ROS中的超氧阴离子含量稍高于对照组,d5差异有统计学意义(P〈0.05)。结论短波紫外线病毒灭活技术处理血小板导致其活化、凋亡增加,HSR下降,ROS生成增加,该灭活技术对血小板体外功能有一定的影响。Objective To investigate the influence on the function related parameters of platelets in vitro after treatment with ultraviolet C light virus inactivation technology.Methods Buffy coat platelet concentrates were resuspended in a mixed solution of PPP and SSP +,adjusted platelet counts about 1 000× 10 9/L. Then the concentrates were exposed to UVC with a dose of 0. 220 J/cm 2.After that the morphology of platelets was observed by scanning electron microscope.Additionally,evaluated the effect on the function related indexes of platelets in vitro at the first,third,fifth and seventh day after UVC irradiated.Results The scanning electron microscopy showed that more irregular shapes and pseudopodiums formation of platelets after UVC irradiated.Platelet counts and HSR of experiment group decreased.MPV and P-LCR increased,compared to the control group,all of these differences were statistically significant( P〈0. 05).The OD values which were used to observe the activity of platelet mitochondrial dehydrogenase by CCK-8 were slightly decreased in experimental group,the differences were statistically significant on the fifth day( P〈0. 05).The platelet activation markers PAC-1 and CD62p were increased after UVC irradiation,as well as apoptosis marker Annexin V,all of these differences were statistically significant( P〈0. 05). The superoxide anion in ROS slightly increased after UVC irradiated,the differences were statistically significant on the fifth day( P 〈0. 05).Conclusion Platelet activation,apoptosis,ROS production increased and HSR decreased after treatment with UVC virus inactivation technology.UVC virus inactivation technology had some effect on the function of platelets in vitro.
