注射性壳聚糖β-甘油磷酸钠凝胶负载富血小板血浆和骨髓基质干细胞促进牙周再生的研究

摘　　要：目的探究注射性壳聚糖β-甘油磷酸钠(chitosan/β-glycerol phosphate disodium salt,C/GP)凝胶负载富血小板血浆(platelet-rich plasma,PRP)和骨髓基质干细胞((bone marrow mesenchymal stem cells,BMSCs)促进牙周再生的效果。方法选取3只健康雄性杂种犬,分别在前磨牙、第一磨牙区制备30颗牙的Ⅱ度根分叉区缺损模型,并随机将30颗牙纳入A组(C/GP)、B组(C/GP+PRP)、C组(C/GP+BMSCs)、D组(C/GP+PRP+BMSCs)和对照组,术中将所有牙周缺损处的组织瓣进行缝合,并在A^D组模型中注入凝胶,对照组则不予处理。采集术后8周时的组织标本,进行X线及组织学观察与评价。结果 B、C、D组与A组和对照组相比均有明显的牙周组织再生(P<0.05),D组与B、C组相比牙周组织再生最明显(P<0.05);A组和对照组仅有少量的牙周组织再生,两组差异不显著(P>0.05)。结论 C/GP负载PRP和BMSCs有助于促进牙周组织的再生,值得进一步研究与利用。Objective To explore the injectable chitosan glycerophosphate（chitosan/-glycerol phosphate disodium salt beta,C/GP）gel load of platelet rich plasma（platelet-rich plasma,PRP）and bone marrow stromal stem cells（bone marrow mesenchymal stem（cells,BMSCs）and promote periodontal regeneration effect.Methods 30 second degree root bifurcation defect mode was made at premolar and first molar area of 3 healthy male dogs.30 premolars were randomly into A group（C/GP）,B group（C/GP＋PRP）,C group（C/GP＋BMSCs）,D group（C/GP＋PRP＋BMSCs）and control group.Intratoperatively,all periodontal defect flap suture,and injected gel in A group,B group,C group and D group.The control group was not treated with.Tissue samples were collected at 8 weeks after operation.X-ray and histological observation and evaluation were carried out.Results The periodontal tissue regeneration of B group,C group and D group were obvious.The periodontal tissue regeneration of A group and control group were few.Conclusion C/GP loaded PRP and BMSCs can promote the regeneration of periodontal tissue.
