摘  要:目的 观察过表达细胞周期蛋白依赖性激酶2A(CDKN2A)基因对食管鳞癌细胞株TE-13增殖、迁移能力的影响.方法 采用脂质体转染法将已构建好的PcDNA3.1_CDNK2A及空载体质粒PcDNA3.1_neo分别转染人食管癌细胞TE-13,分为实验组(PcDNA3.1_CDNK2A_ TE-13),阴性对照组(PcDNA3.1_neo-TE-13)及空白对照组(只添加转染试剂).采用实时定量聚合酶链反应(Real-time PCR)检测转染前后CDKN2A mRNA表达水平;应用噻唑蓝(MTT)法检测转染后细胞的增殖能力;采用Transwell实验检测细胞的迁移能力.结果 实验组、阴性对照组及空白对照组CDNK2A mRNA相对表达水平分别为0.45 ±0.10、0.48±0.12及2.12±0.24,实验组显著高于阴性对照和空白对照组(F=200.500,P=0.000).实验组、阴性对照组及空白对照组吸光度值分别为0.71 ±0.08、1.05±0.09和0.97±0.08,差异有统计学意义(F=27.220,P=0.000).Tanswell结果显示实验组、阴性对照组及空白对照组穿过生物膜细胞分别为(156.8±23.1)、(166.9 ±28.4)和(162.1 ±30.3)个,差异无统计学意义(F=0.200,p=0.818).结论 CDKN2A基因表达能够显著抑制食管癌细胞的增殖,但对其迁移能力无明显影响.Objective The aim of this study was to evaluate the overexpression cyclin-dependent kinase 2A (CDKN2A) gene in human esophageal cancer cell TE-13 and its function for proliferation.Methods Human esophageal cancer cell TE-13 was transfected with PcDNA3.1_ CDNK2A and PcDNA3.l_neo as experiment and negative control group.The real-time quautitative polymerase chain reaction (Real-time PCR) was used to assess the CDNK2 expression after transfection.MMT assay and Transwell assay were used to examine the cell proliferation and invasion ability.Results CDNK2A mRNA relative expression were 0.45 ±0.10,0.48 ±0.12 and 2.12 ±0.24 respectively with statistical difference in experiment group,negative group and blank control group respectively,which indicating CDNK2A mRNA can be overexpressed by transfecting PcDNA3.1_CDNK2 (F =200.500,P =0.000);D490 value were 0.71 ±0.08,1.05 ±0.09 and 0.97 ±0.08 respectively with statistical difference (F =27.220,P =0.000).Transwell results showed the pass through biomembrane cells were (156.8 ±23.1),(166.9 ±28.4) and (162.1 ± 30.3) cells without statistical difference (F =0.200,P =0.818).Conclusion Overexpression CDNK2A can inhibit human esophageal cancer cell TE-13 proliferation but not affect its invasion ability.
