摘 要:目的观察薯蓣皂苷元(Dio)对人多发性骨髓瘤RPMI8226细胞株增殖、迁移及侵袭的影响,并探讨其可能作用机制。方法取对数期RPMI8226细胞分1、2、3、4、5组,1、2、3、4、组分别加入10、20、30、40μmol/L浓度的Dio、5组为MM细胞置于单纯培养基中,不做任何处理。分别于培养24、48、72 h在490 nm波长的酶标仪中检测各孔光密度值(OD值),观察细胞增殖抑制情况。将RPMI8226细胞分为高浓度组、中浓度组、低浓度组、硼替佐米(BTZ)浓度组、对照组,高、中、低浓度组分别加入16、12、8μL的40、30、20μmol/L的Dio,BTZ组加入10 nmol/L的BTZ 2μL,对照组不做任何处理,采用细胞迁移实验观察各组细胞迁移能力,采用细胞侵袭实验观察各组细胞侵袭能力,采用ELISA检测各组细胞上清白细胞介素6(IL-6)、血管内皮生长因子(VEGF)、IL-8蛋白。结果随培养时间增加,1、2、3、4组细胞OD值增加(P均<0.05)。随作用浓度增加,细胞OD值增加(P<0.05)。与对照组比较,其余各组迁移、侵袭细胞数减少(P均<0.05);与BTZ组比较,高、中、低Dio浓度迁移、侵袭细胞数减少(P均<0.05).与对照组比较,其余组IL-6、VEGF、IL-8相对表达量减少(P均<0.05);与BTZ组比较,高、中、低Dio组IL-6、VEGF、IL-8相对表达量减少(P均<0.05)。结论 Dio可抑制RPMI8226细胞的增殖、迁移、侵袭,其机制可能与其下调细胞IL-6、VEGF、IL-8表达相关。Objective To observe the effects of diosgenin(Dio) on the proliferation,migration,and invasion of human multiple myeloma cell line RPMI8226 in vitro,and to explore its mechanism. Methods The RPMI8226 cells in the logarithmic phase were divided into 5 groups: 10,20,30,40 μmol/L Dio groups(groups 1,2,3,and 4) and the normal group(group 5). The OD value of each group was detected by MTT at 24,48,and 72 h,and the inhibition rate of cell proliferation was calculated. Then the RPMI8226 cells in the logarithmic phase were divided into the high-dose,mediumdose and low-dose Dio groups,Bortezomib group(BTZ),which were added with 16,12,and 8 u L Dio with the concentrations of 40,30,and 20 μmol/L,2 μL BTZ(10 nmol/L),and the control group without any treatment. Transwell was used to detect the migration and invasion of cells,and the changes of IL-6,vascular endothelial growth factor(VEGF) and IL-8 in the supernatant of each group were detected by ELISA. Results The OD values increased in the groups 1,2,3,and 4 in a time-and dose-dependent manner(all P〈0. 05). Compared with the control group,the migration and invasion cells,and the levels of IL-6,VEGF,and IL-8 of the other groups decreased(all P〈0. 05). Compared with the BTZ group,the migration and invasion cells and the expression levels of IL-6,VEGF,and IL-8 in the high-dose,medium-dose and low-dose Dio groups decreased significantly(all P〈0. 05). Conclusion Dio can inhibit the proliferation,migration,and invasion of RPMI8226 cells by down-regulating the expression of IL-6,VEGF,and IL-8.
