摘　　要：目的:研究抗胃泌素抗体及其联合化疗药的体外抑瘤作用。方法:Western blot法筛选可自分泌胃泌素并表达胃泌素受体的肿瘤细胞系;MTT法研究胃泌素、化疗药(顺铂、氟尿嘧啶)、抗胃泌素抗体及其联合化疗药对其生长的作用;Western blot法研究上述联合抑瘤方法的作用机制。结果:人胃癌细胞系SGC7901可自分泌胃泌素并表达胃泌素受体;胃泌素可显著促进其生长,顺铂、氟尿嘧啶、抗胃泌素抗体均可显著抑制其生长;抗胃泌素抗体联合化疗药具有协同抑瘤作用,该抗体可通过抑制肿瘤细胞内耐药蛋白(GST-π、TS)的表达提高肿瘤细胞对化疗药(顺铂、氟尿嘧啶)的敏感性。结论:抗胃泌素抗体及其联合化疗药具有较好的抑制人胃癌细胞系SGC7901体外增殖的作用。Objective：To study the in vitro anti-tumor effect of anti-gastrin antibody and its combination with chemical drugs.Methods： The tumor cell lines,which can excrete gastrin and express corresponding receptor,were screened by Western blot.The effects of gastrins,chemotherapeutic drugs（e.g.cisplatin,fluorouracil）,anti-gastrin antibody and its combination with the chemotherapeutic drugs on the growth of the cell line were identified by MTT,respectively.The anti-tumor mechanism of the combinational antibody-drug treatment in the cell line was studied by Western blot.Results：The human gastric tumor cell line of SGC7901 could excrete gastrin and express corresponding receptor.Gastrin could significantly promote the cell growth.Cisplatin,fluorouracil and the antibody all could inhibit the growth of the cell line significantly.The combinational antibody-drug treatment showed synergic anti-tumor effect.The antibody could promote the sensitivity of the cell to the chemotherapeutic drugs（e.g.cisplatin,fluorouracil）by inhibiting the expression of drug resistance proteins（e.g.GST-π,TS）.Conclusion：The anti-gastrin antibody and its combination with chemotherapeutic drugs have a good effect in inhibiting the proliferation of SGC7901 human gastric tumor cell line in vitro.
