摘 要:目的:探讨肿瘤坏死因子-α(TNF-α)对人牙周膜干细胞(PDLSCs)成骨作用的影响,以及其中长链非编码(LncRNA)的差异表达情况。方法:分离培养PDLSCs,对照组细胞用成骨诱导液培养,实验组细胞用成骨诱导液+10 ng/ml TNF-α培养。通过茜素红染色、RT-PCR和Western bolt检测两组细胞骨向分化,用高通量测序技术探究LncRNA的差异表达情况。结果:在TNF-α作用下,PDLSCs的骨向分化能力减弱;测序结果发现差异表达显著的长链非编码RNA上调的有57条,下调的有26条。结论:LncRNA可能与PDLSCs骨向分化的调控密切相关。Objective: To study the osteogenic differentiation of periodontal ligament stem cells(PDLSCs) regulated by tumor necrosis factor-α(TNF-α) and the differential expression of long noncoding RNA(LncRNA). Methods: PDLSCs were cultured by osteogenic induction medium with and without TNF-α respectively. The osteogenic differentiation ability of PDLSCs were tested by Alizarin red staining, RT-PCR and Western blot. The differential expression of LncRNA was explored by high-throughput sequencing. Results: TNF-α inhibited osteogenic differentiation potential of PDLSCs. There were 57 up-regulated and 26 down-regulated LncRNAs in PDLSCs treated by TNF-α. Conclusion: LncRNA may be closely related to the regulation of osteogenic differentiation of PDLSCs.
