摘  要:目的优化黄芪茯苓组方,探讨其提高机体缺氧耐力及其作用机制。方法采用常压密闭缺氧实验,以0.5%的羧甲基纤维素为空白对照,黄芪茯苓原方为阳性对照,观察不同剂量的新黄芪茯苓对小鼠常压缺氧的保护作用。结果与空白对照组相比较,黄芪茯苓原方组、新黄芪茯苓低、中、高剂量组小鼠的缺氧生存时间明显延长(P<0.05),而且新黄芪茯苓各组与黄芪茯苓原方组之间没有统计学差异;新黄芪茯苓可使小鼠血清中总抗氧化能力(T-AOC)、过氧化氢酶(CAT)、总超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活力升高,丙二醛(MDA)含量下降,心肌和脑组织中Na^+K^+-ATP酶和Ca^(2+)Mg^(2+)-ATP酶的活力提高。结论新黄芪茯苓可延长小鼠常压耐缺氧时间,改善缺氧状态,减轻自由基对小鼠的损伤,改善心肌和脑组织能量代谢,对抗缺氧损伤,这可能是其对抗缺氧的机制之一。Objective To optimize the Huangqi-Fuling prescription(HF), and investigate its anti-hypoxic action and the mechanism involved. Methods A total of 52 mice were used to observe the protective effect of HF on normobaric hypoxia, with 0.5% carboxymethyl cellulose as the blank control group. Results The hypoxia tolerance survival time of mice in HF low, medium and high dose groups was significantly prolonged, compared with the blank control group. But there was no statistically significant difference among the HF groups. HF could increase the activities of T-AOC, CAT, SOD and GSH, decrease the content of MDA in the serum, and increase the activities of Na+K+-ATP and Ca2+Mg2+-ATP enzymes in myocardium and brain tissues. Conclusion HF can prolong the survival time of hypoxia tolerance and improve the hypoxia state of mice. The anti-hypoxia mechanisms of HF may be related to reduced damage of free radicals, improving energy metabolism in myocardium and brain tissues, and protecting the organism from hypoxia injury.
