摘  要:肿瘤细胞的体外培养对研究肿瘤复杂的病理生理机制做出了巨大的贡献。近年来,三维细胞培养广泛应用于生物医学研究的各个领域,尤其是在肿瘤研究中发展迅速。肿瘤细胞的三维培养可以更准确地在体外模拟体内肿瘤真实的生长环境,肿瘤细胞在各种三维培养装置中呈立体球状生长,表现出与体内肿瘤相似的形态学和细胞生物学特性。应用三维细胞培养技术在体外构建肿瘤模型逐渐成为连接体外肿瘤细胞二维培养和体内肿瘤动物模型之间的桥梁,目前已广泛应用于研究肿瘤生物学行为、肿瘤微环境、肿瘤干细胞、肿瘤血管生成和抗肿瘤药物筛选等方面。本文就近年来三维细胞培养的进展及其在肿瘤研究中的应用作一综述。In vitro culture of tumor cell lines has made great contributions to the study of the complex pathophysiology of tumor. Three-dimensional cell culture has been widely used in various fields of biomedical research over the past few years, especially in cancer research. Tumor cells cultured in a three-dimensional condition can more accurately simulate the growth environment of tumor in vivo. Tumor cells grow into spheroid in various three-dimensional culture devices, showing the morphological and cellular biological characteristics similar to solid tumor. The three-dimensional cell culture model in vitro has gradually become a bridge between monolayer culture in vitro and animal models in vivo, and it has been widely applied in the study of tumor biological behavior, tumor microenvironment, tumor stem cells, tumor angiogenesis, anti-tumor drug screening and so on. This paper reviews the progress in three-dimensional cell culture and its application in tumor research in recent years.
