摘 要:以中条山的野生复叶葡萄籽为材料,采用香草醛比色法和超声波辅助法,采用单因素试验研究了原花青素提取率影响因子,通过正交优化和多重比较得出优化方案。结果表明:提取液用2.0mL测定更为合适;原花青素含量低的提取液颜色深,相反,则颜色浅。同时在单因素的基础上,经过正交优化和多重比较获得最佳工艺条件为:料液比1∶55g·mL^(-1),超声波功率60W,温度60℃,时间55min。在此工艺条件下,原花青素的提取率为1.983%。The seeds of wild grape in Zhongtiao mountain were used as the materials,the determination method was vanillin colorimetry,and the extraction method was ultrasonic-assisted method.The concentration of proanthocyanidin in the extract was obtained from the standard curve of proanthocyanidin standard product.The factors affecting the extraction rate of proanthocyanidins were studied by single factor experiments,and the optimization combination was obtained by orthogonal experiment and multiple comparison.The results showed that 2.0 mL was more suitable for the determination of the extract.The extract with low proanthocyanidin content was dark in color,whereas the extract with high proanthocyanidin content was light in color.And the optimum conditions were obtained by orthogonal optimization experiment and multiple comparison,solid-liquid ratio 1∶55 g· mL^-1,ultrasonic power 60 W,temperature 60 ℃,time 55 minutes.Under these conditions,the extraction rate of proanthocyanidins was 1.983%.
