摘 要:目的:观察葛根枳椇子栀子胶囊对血液乙醇浓度,肝组织酒精代谢关键酶、抗氧化酶活性及血清中肝毒性生物标志物含量的影响,探讨其解酒护肝作用机制。方法:采用顶空-气相色谱法(HS-GC)检测大鼠酒后不同时间血液乙醇浓度;检测急性酒精中毒小鼠肝组织中乙醇脱氢酶(ADH)、乙醛脱氢酶(ALDH)、超氧化物歧化酶(SOD)活性和丙二醛(MDA)的含量;检测急性酒精性肝损伤小鼠血清谷丙转氨酶(ALT)、谷草转氨酶(AST)水平。结果:葛根枳椇子栀子胶囊低剂量组大鼠酒后30 min、中剂量组酒后60 min、高剂量组酒后30、60、90、120、180 min血液乙醇浓度均显著低于模型组(P<0.01或P<0.05)。葛根枳椇子栀子胶囊中、高剂量组小鼠肝组织ADH活性显著高于模型组(P<0.05),高剂量组肝组织ALDH和SOD活性显著高于模型组(P<0.05或P<0.01),而MDA含量显著低于模型组(P<0.05)。葛根枳椇子栀子胶囊中、高剂量组小鼠血清ALT含量均低于模型组,但差异无统计学意义(P>0.05),而血清AST含量显著低于模型组(P<0.05)。结论:葛根枳椇子栀子胶囊具有明显的解酒护肝作用,这可能与其增强肝乙醇代谢关键酶及抗氧化酶活性,加速乙醇在体内代谢,减少自由基及脂质过氧化物的产生,保护肝细胞膜完整性相关。Objective: To investigate the effect of Gegen Zhijuzi Zhizi capsule on anti-alcohol and hepatoprotection. Methods: The level of blood alcohol in rats was measured by HS-GC, and the content of liver ADH,ALDH, SOD and MDA in mice were also determined. The levels of serum ALT, AST in acute alcoholic liver injury mice were detected. Results: The level of blood alcohol in rats of the low-dose group after 30 min, middle-dose group after 60 min and high-dose group after 30, 60, 90, 120, 180 min decreased significantly than that in the model group(P〈0.01 or P〈0.05). The activity of liver ADH increased markedly in the middle-dose group and high-dose group(P〈0.05), and the activities of ALDH and SOD in liver increased markedly(P〈0.05 or P〈0.01), but the content of MDA reduced in the high-dose group of Gegen Zhijuzi Zhizi capsule(P〈0.05).Besides, the level of serum ALT in the middle-dose group and high-dose group was lower than that in model group, but the differences were not significant(P 0.05). The level of serum AST was significantly reduced as compared with model group in acute alcoholic liver injury mice(P〈0.05). Conclusion: The Gegen Zhijuzi Zhizi capsule has obvious anti-alcohol and hepatoprotective effects, which may be related to the activity enhancement of liver ethanol metabolism key enzymes and antioxidant enzyme, acceleration of ethanol metabolism in the body,decrease of the free radicals and lipid peroxides production, and protection of liver cell membrane integrity.
