摘 要:目的观察黄酒对二硝基氟苯诱导的小鼠慢性变应性接触性皮炎(ACD)的影响。方法 30只小鼠随机分成模型组、黄酒组和空白组,每组10只。模型组和黄酒组用2,4-二硝基氟苯反复诱导建立慢性ACD模型。在第4次激发前1天分别用纯水和黄酒灌胃,1次/d。第4次激发后48h测量各组小鼠左右耳厚度差,计数炎症细胞,用ELISA法检测血清IL-4、IL-17和IFN-γ水平,RT-PCR法检测耳组织中IL-4、IL-17和IFN-γmRNA水平。结果 4次激发后黄酒组的耳厚度差、炎症细胞计数、血清IL-17蛋白水平和耳组织内IL-17 mRNA相对表达水平均明显高于模型组(P<0.05),但黄酒组血清中IL-4和IFN-γ水平和耳组织中IL-4和IFN-γmRNA相对表达水平与模型组差异无统计学意义(P均>0.05)。结论黄酒可能通过上调IL-17水平加重慢性ACD小鼠模型的炎症反应。Objective To observe the effects of rice wine on chronic allergic contact dermatitis(ACD)mice model. Methods Thirty BALB/c mice were randomly divided into model group, rice group and control group with 10 mice in each. Model group and rice wine group were built by repeating treatment of 2,4-dinitrofluorobenzene(DNFB). One day before the fourth elicitation, those mice were fed respectively with pure water and rice wine by gavage once a day. 48 hours after the fourth elicitation, the ear thickness difference, the number of infiltrating inflammatory cells were measured. The levels of interleukin 4(IL-4), interleukin 17(IL-17)and interferonγ(IFN-γ)in the serum were detected by ELISA, and the levels of IL-4, IL-17 and IFN-γ mRNA in the ear tissue were checked by reverse transcription-polymerase chain reaction(RT-PCR). Results After the fourth elicitation, the ear thickness difference, the number of infiltrating inflammatory cells, the levels of IL-17 in the serum and IL-17 mRNA in the ear tissue in the rice wine group were significantly higher than those in the model group(P〈0.05,respectively),but no significant differences were detected on the levels of IL-4 and IFN-γ in the serum and the levels of IL-4 and IFN-γ mRNA in the ear tissue(all P〉0.05). Conclusion Rice wine can exacerbate the inflammatory reaction of chronic ACD model in mice through up-regulation of the IL-17 expression.
