摘 要:目的探讨microRNA-145(miR-145)调控Catenin-δ1表达与结直肠癌侵袭和转移的关系。方法应用实时荧光定量PCR和显色原位杂交技术检测36例结直肠癌组织和配对正常肠粘膜组织中miR-145的表达;利用LipofectamineTM2000试剂将miR-145模拟物瞬时转染SW620肠癌细胞株,通过RT-PCR和Westernblot分析Catenin-δ1及Fascin-1表达情况;CCK-8试剂盒和Transwell小室检测各组细胞转染后增殖、侵袭和迁移能力的变化。结果miR-145在结直肠癌组织中的表达低于配对正常肠粘膜组织,且表达水平与淋巴结转移相关(P<0.05)。体外细胞实验显示,转染miR-145模拟物可显著降低SW620肠癌细胞株Catenin-δ1和Fascin-1蛋白表达水平,并抑制肿瘤细胞增殖、侵袭和迁移。结论miR-145低表达与结直肠癌侵袭和转移相关,其机制可能与调控Catenin-δ1的表达水平相关。Objective To investigate the role of micro RNA-145(miR-145)in colorectal cancer(CRC)invasion and metastasis. Methods The expression levels of miR-145 were determined by real-time quantitative PCR and in situ hybridization(ISH)in 36 CRC tissues and matched adjacent normal mucosa. miR-145 mimics and siRNA were transiently transfected into human colorectal cancer cells to up-regulate or down-regulate gene expression,and their effects on cell migration and invasion were analyzed by Transwell assay. Cell proliferation was measured using the Cell Counting kit-8. The expression of Catenin-δ1 and Fascin-1 was examined by Western blot. Results miR-145 was down-regulated in CRC tissues,and low expression of miR-145 was significantly associated with lymph node metastasis(P〈0.05). Transfection of mi R-145 mimics inhibited proliferation,invasion and migration in SW620 cells. Furthermore,up-regulation of miR-145 inhibited the expression levels of Catenin-δ1 and Fascin-1. Conclusion miR-145 played an important role in mediating invasive and metastatic behavior in CRC,probably through regulating the expression of Catenin-δ1.
