摘  要:目的建立一种高效液相色谱法同时测定四物汤中5-羟甲基糠醛、川芎嗪、芍药苷、阿魏酸四种活性成分的含量。方法采用Phenomenex luna C18(250mm×4.6mm,5.0μm)色谱柱,流动相:乙腈(A)-0.05%甲酸水溶液(B),线性梯度洗脱(0~5min,A相5%;5~10min,A相5%→10%;10~20min,A相10%→20%;20~40min,A相20%→30%;40→46min,A相30%→5%);流速:1.0mL·min^(-1);检测波长:230nm和282nm。柱温:35℃。结果 5-羟甲基糠醛、川芎嗪、芍药苷、阿魏酸分别在0.0215~0.43mg·mL^(-1)、0.021~0.42mg·mL^(-1)、0.0195~0.39mg·mL^(-1)、0.023~0.46mg·mL^(-1)范围内呈良好线性关系。四物汤中5-羟甲基糠醛、川芎嗪、芍药苷、阿魏酸四种有效成分的含量分别为0.0681%、未检出、0.250%、0.0358%。结论该方法操作简便,结果准确可靠。OBJECTIVE To establish a method for simultaneous determination of 5-HMF,tetramethyl-pyrazine,paeoniflorin and ferulic acid in Siwu Decoction. METHODS A Phenomenex Luna C18 column(4. 6 mm × 250 mm,5μm) was used. The mobile phase consisted of acetonitrile(A)-water containing 0. 05% formic acid(B) with linear gradient elution(0 ~ 5 min,A: 5%; 5 ~ 10 min,A: 5% →10%; 10 ~ 20 min,A: 10% →20%; 20 ~ 40 min,A: 20% →30%; 40→46 min,A: 30% →5%). The flow rate was 1. 0 mL · min^(-1). The detection wavelength was 230 nm and 282 nm. The column temperature was controlled at 35℃. RESULTS 5-HMF,tetramethylpyrazine,paeoniflorin and ferulic acid showed good linearity in the range of 0. 0215 ~ 0. 43 mg · mL^(-1),0. 021 ~ 0. 42 mg·mL^(-1),0. 0195 ~0. 39 mg·mL^(-1) and 0. 023 ~ 0. 46 mg·mL^(-1),respectively. The contents of 5-HMF,tetramethylpyrazine,paeon-iflorin and ferulic acid in Siwu Decoction were 0. 0681%,not be detected,0. 249% and 0. 0357%,respectively. CONCLUSION The method has simple operation and high accuracy.
