结直肠癌miR-200a与miR-92a表达水平及其与临床病理特征的关系

摘　　要：目的研究miR-200a、miR-92a在结直肠癌中的表达水平及其与患者临床病理特征的关系。方法选取2011年2月~2012年8月在新疆医科大学附属肿瘤医院接受手术治疗的结直肠癌患者60例,采集结直肠癌组织与癌旁组织标本各60例。采用原位分子杂交技术检测上述标本中miR-200a表达阳性率,采用实时荧光定量PCR法检测上述标本中miR-92a表达情况。分析miR-200a及miR-92a表达情况与结直肠癌患者的临床病理特征相关性。随访5年,根据生存状态分为存活组与死亡组,比较两组患者miR-200a、miR-92a表达情况差异。结果结直肠癌组织中miR-200a表达阳性率及miR-92a相对表达明显高于癌旁组织(均P<0.05)。中高分化、TNMⅠ~Ⅱ期结直肠癌患者miR-200a表达阳性率明显低于低分化、TNMⅢ~Ⅳ期的结直肠癌患者,分别为40.00%(18/45)比73.33%(11/15)、58.06%(18/31)比86.21%(25/29),差异均有统计学意义(均P<0.05)。而TNMⅠ~Ⅱ期、无淋巴结转移结直肠癌患者miR-92a相对表达量均明显低于TNMⅢ~Ⅳ期、有淋巴结转移患者(均P<0.05)。随访5年,生存组21例,死亡组39例。存活组miR-200a表达阳性率及miR-92a相对表达量均明显低于死亡组,差异均有统计学意义(均P<0.05)。结论结直肠癌组织的miR-200a、miR-92a高表达,与病情严重程度及预后密切相关。Objective To analyze the expression of mi R-200 a and mi R-92 a in colorectal cancer and the relation of clinicopathological features. Methods The 60 patients with colorectal cancers in Affiliated Tumor Hospital, Xinjiang Medical University for surgical treatment from February 2011 to August 2012 were collected. And the 60 colorectal cancer tissue specimens and paracancerous tissues were achieved in patients with colorectal cancer respectively. The positive expression rates of mi R-200 a in colorectal cancer tissues and paracancerous tissues were detected by in situ hybridization technique. The expression levels of mi R-92 a in colorectal cancer tissues and paracancerous tissues were detected by real-time PCR. The relativities of the expression of mi R-92 a and mi R-200 a and the clinicopathological features were analyzed. Follow-up 5 years, all patients with colorectal cancers were divided into survival group and death group according to the survival status of patients. The expression levels of mi R-92 a and mi R-200 a were compared between two groups. Results The positive rate of mi R-200 a and the expression level of mi R-92 a in colorectal cancer tissues were significantly higher than in paracancerous tissues（all P〈0.05）. The positive expression rates of mi R-200 a in the colorectal cancer patients of the middle and high degree of differentiation and TNM stage Ⅰ-Ⅱ were significantly lower than those of the low degree of differentiation and TNM stage Ⅲ-Ⅳ, which were 40.00%（18/45） vs.73.33%（11/15）, 58.06%（18/31） vs. 86.21%（25/29）（all P〈0.05）. The relative expressions of mi R-92 a in colorectal cancer patients of the TNM stage Ⅰ-Ⅱ and without lymph node metastasis were significantly lower than those of the TNM stage Ⅲ-Ⅳ and lymph node metastasis, the differences were statistically significant（all P〈0.05）.
