摘  要:目的探讨DMT1抑制剂ebselen是否通过抑制细胞内铁离子内流,发挥抑制Aβ产生的作用。方法培养APPsw细胞,MTT法检测ebselen对细胞活力的影响,分别给予FeSO_4或ebselen处理细胞,ELISA检测Aβ1-42水平,免疫荧光检测DMT1和BACE1的表达,以及荧光分光光度计检测ebselen对二价铁离子转运的影响。结果 ebselen作为DMT1抑制剂,能抑制二价铁离子的细胞内转运,降低DMT1和BACE1的表达水平,抑制铁诱导的APPsw细胞内Aβ1-42水平。结论 ebselen抑制铁诱导的APPsw细胞内Aβ1-42水平,从而抑制Aβ的产生。Objective To study whether ebselen, as DMT1 inhibitor, plays an important role in inhibiting the production of Aβ by inhibiting intracellular iron ion flow. Methods APPsw cells were treated by FeSO4 and(or) ebselen. The cell viability was determined by MTT method, Aβ1-42 level was detected by ELISA method. The expression of DMT1 and BACE1 was determined by immunofluorescence method, and the ebselen affecting bivalent iron ion transportion was determined by fluorescence spectrophotometer method. Results ebselen inhibited the intracellular transport of divalent iron ions, decreased the expression level of DMT1 and BACE1, and inhibited the level of Aβ1-42 in APPsw cells. Conclusion ebselen inhibits the level of Aβ 1-42 in APPsw cells as a DMT1 inhibitor.
