摘  要:目的观察长链非编码RNA HOXD-AS1对骨肉瘤细胞增殖的影响及可能机制。方法应用Western blot与real-time PCR方法检测长链非编码RNA HOXD-AS1与RUNX3在骨肉瘤组织及癌旁组织中的表达,CCK-8法检测过表达长链非编码RNA HOXD-AS1后对骨肉瘤细胞增殖活力的影响以及RUNX3的表达变化,过表达RUNX3后检测长链非编码RNA HOXD-AS1对骨肉瘤细胞增殖活力的影响。结果长链非编码RNA HOXD-AS1在骨肉瘤组织中表达水平高于癌旁正常组织,而RUNX3在骨肉瘤组织中的表达低于癌旁正常组织,过表达长链非编码RNA HOXD-AS1表达后骨肉瘤细胞增殖活力显著增强,RUNX3的表达水平减少,过表达RUNX3后过表达长链非编码RNA HOXD-AS1,长链非编码RNA HOXD-AS1对骨肉瘤细胞的促增殖抑制作用减弱。结论长链非编码RNA HOXD-AS1促进骨肉瘤细胞增殖与RUNX3相关。Objective To observe the effect of lncRNA HOXD-AS1 on the proliferation and possible mechanism of osteosarcoma cells. Methods Western blot and real-time RT-PCR were used to detect the expressions of lncRNA HOXD-AS1 and RUNX3 in osteosarcoma tissues and adjacent normal tissues. The proliferation ability of osteosarcoma cells was detected by CCK-8. Results The expression level of lncRNA HOXD-AS1 was significantly increased in osteosarcoma tissues than in the adjacent normal tissues, but decreased for RUNX3 expression level. Overexpression of lncRNA HOXD-AS1 could promote the cells proliferation and inhibit the expression of RUNX3 in osteosarcoma cells. Over-expression of RUNX3 could possibly reverse the acceleration of the cellular viability after lncRNA HOXD-AS1 over-expression. Conclusion The promotion of lncRNA HOXD-AS1 on osteosarcoma cells might be related to RUNX3.
