摘  要:目的:观察表没食子儿茶素没食子酸酯(Epigallocatechin gallate,EGCG)对2型糖尿病大鼠认知功能的影响。方法:雄性Wistar大鼠,除对照组外均采用链脲菌素联合高脂饮食复制2型糖尿病模型。造模成功后,随机分成模型组,二甲双胍组(140 mg·kg-1·d-1,灌胃),EGCG低、中、高剂量组(分别为10、20、40 mg·kg-1·d-1,灌胃)。给药4周后,采用Morris水迷宫法进行行为学检测,测定大鼠空腹血糖(FBG)、血甘油三酯(TG)、总胆固醇(TC)、海马丙二醛(MDA)、一氧化氮(NO)水平及超氧化物歧化酶(SOD)活性。结果:与模型组比较,EGCG高剂量组寻台时间缩短,穿台次数增多(P〈0.05);FBG、TG、TC、海马MDA和NO含量显著降低,SOD活性显著增加(P〈0.05)。结论:EGCG可改善2型糖尿病大鼠认知障碍,可能与其降低动物体内血糖、提高抗氧化能力有关。Objective:To observe the effects of epigallocatechin gallate(EGCG)on cognitive function of type 2 diabetes mellitus rats. Methods:Male Wistar rats were randomly divided into control group and test group. The test group was used by the combination of streptozotocin and hight-fat diet to establish the model of T2 DM. Then the model of T2 DM rats were randomly divided into model group,metformin group,EGCG low dose group,EGCG middle dose group and EGCG high dose group. Metformin group was treated with metformin(140 mg·kg-1)by gavage once daily. EGCG groups were treated with EGCG(10,20,40 mg·kg-1)by gavage once daily. After 4 weeks,the Morris water maze task was used for behavior test.The fasting blood glucose(FBG),blood fat(TG),total cholesterol(TC),nitric oxide(NO),superoxide dismutase(SOD)and malonaldehyde(MDA)in hippocampus were detected. Results:Compared with model group,EGCG high dose group decreased the swimming time to platform,had more times crossing the hidden platform(P〈0.05),decreased the level of FBG,TG,TC,and decreased the level of NO and MDA in hippocampus,improved the activity of SOD(P〈0.05). Conclusion:EGCG could improve the cognitive disorder of type 2 diabetes mellitus rats. The mechamism might be related to decrease blood glucose and increase the ability of anti-oxidative ability.
