摘  要:目的探讨下调EZH2基因对胃癌细胞株SGC7901的增殖作用,研究EZH2基因在胃癌发生发展中的作用。方法针对EZH2基因序列设计合成小干扰RNA片段(si RNA)及无效序列片段,将EZH2 si RNA转染至胃癌细胞株SGC7901中作为干扰组,以转染无效序列片段作为阴性对照组,以未作任何处理组作为空白对照组,采用荧光定量PCR(q RT-PCR)检测EZH2基因下调后EZH2 m RNA的表达,采用Western blot法检测EZH2蛋白表达量,采用CCK-8法检测细胞增殖的活性。结果成功转染EZH2 si RNA至胃癌SGC7901细胞,干扰组的EZH2m RNA和蛋白相对表达量较阴性对照组及空白对照组下降,且干扰组细胞增殖受到抑制。结论下调EZH2基因的表达,使胃癌细胞增殖受到抑制,说明EZH2基因在胃癌细胞的增殖发展进程中有很重要的意义,为深入研究胃癌基因治疗提供一定的理论依据。Objective To investigate the effect of EZH2 downregulation on cell proliferation in human gastric cancer(GC) cell line SGC7901, to determine the effect of EZH2 gene in the occurrence and development of gastric cancer. Meth Method The EZH2 gene sequence targeted siRNA were designed and synthesized, with a scramble sequence as negative control,besides, a blank control without any treatment was used. The expression of EZH2 mRNA level was detected by qRT-PCR after downregulation of EZH2, Western blot was used to detect the expression of EZH2 protein, and CCK-8 assay was utilized to observe the proliferative activity of SGC7901 cells. Result EZH2 siRNA were successfully transfected to gastric cancer SGC7901 cells, the expression of EZH2 mRNA and protein level were significantly suppressed by siRNA compared with negative control and blank control, furthermore, the cell proliferation was inhibited in interference group. Con Conclusion Downregulation of EZH2 gene inhibits the proliferation of gastric cancer cells, suggesting that EZH2 gene is essentially involved in the development and progression of gastric cancer cells, which provides an important theoretical basis for gene therapy of gastric cancer.
