母婴分离方法制备产后抑郁大鼠模型的行为学及海马病理变化的观察  被引量:1

摘  要:目的观察并验证母婴分离方法制备的产后抑郁模型大鼠的行为学与海马病理学表现。方法随机选取16R大鼠进行为期14d的母婴分离,另4只产后母鼠作为空白对照不作任何分离。造模结束后对母鼠行旷场实验评价,根据其评分应用K-聚类分析进行分类,造模1周后对20只大鼠行为学评价,造模5周后取大脑海马观察HE染色结果。结果聚类分析结果得出正常组9只(包括4只空白对照大鼠),产后抑郁组11只。与正常组比较,造模1周后产后抑郁组大鼠体重变化程度明显增加,水平及垂直分数明显降低,悬尾实验不动时间明显增加,十字迷宫开臂停留时间及进入开闭臂总次数明显降低,并且闭臂停留时间明显增加。HE染色显示产后抑郁组海马神经元细胞损伤。结论经过2周母婴分离的母鼠有抑郁表现的概率为68.75%,此模型有必要进一步的探索实验。Objective To observe and verified the behavioral and hippocampal pathological features of postpartum depression model rats prepared by maternal,and infant separation methods. Methods A total of 16 rats were randomly selected for 14d separation, and the other 4 rats served as controls without any separation. The day of birth was considered as Day 0. Maternal separation was carried out daily 9: 00-12:00 during postnatal days 1-14, and pups were placed into a clean cage that near to their home cages. All mothers were subjected to open field test ( OFT ) at Day 14 and Day 21, elevated plus maze test (EPM) at Day 22, forced swing test at (FST) Day 24 and tail suspension test (TST) at Day 25. Hippocampi were HE stained and observed under light microscope at Day 50. The OFT scores of Day 14 including that crossing score, vertical score, average movement speed, total moving distance and center staying time were used as factors for the K-central clustering analysis to separate 20 rats into 2 groups. Results There are 11 in 16 separated mothers were picked out by K-central clustering analysis which were defined as postpartum depression model group ( PPD group ) . While the other 9 mother ( including 4 mother rats in blank group without any treatment ) rats were defined as normal group. Rats in PPD group had significantly weight changes ( P〈0.05 ) than that in normal group. And they had decreased rearing and crossing scores in OFT (P〈0.05) , increased immobility time in TST ( P〈0.05 ) than that in normal group. Mother rats in PPD group spent more time in close arm and their entries into each arm significantly decreased than that in normal group. Five weeks after separation, hippocampus neuron in PPD group were significantly reduced and many of their shape changed into cone and polygon, with nuclear condensation, while those in normal group shows integrity structure in good arrangement with clear and homogeneous nucleus staining. Conclusion The probability of depression is 68.75%
