摘  要:目的研究来那度胺在体内外对人宫颈癌SiHa细胞及其小鼠移植肿瘤的作用,阐明来那度胺对肿瘤的抑制作用及机制。方法 CCK法研究体外来那度胺及DMSO对SiHa细胞生长的影响。建立小鼠人宫颈癌SiHa移植模型,分为实验组及对照组各6只。实验组小鼠每日给予腹腔内注射来那度胺(50mg/kg),对照组小鼠每日给予腹腔内注射1%DMSO,共计21天。3周后处死小鼠取肿瘤组织,比较两组肿瘤组织体积,重量,病理组织学改变,肿瘤组织内血管生成情况及两组小鼠肿瘤转移情况。结果来那度胺在体外对SiHa细胞生长无影响(P>0.05);来那度胺可抑制小鼠移植肿瘤的生长,实验组小鼠移植肿瘤体积和重量低于对照组,差异具有统计学意义(P<0.05);实验组小鼠肿瘤组织内血管生成数量低于对照组,差异具有统计学意义(P<0.05);对照组小鼠出现肺转移灶,而实验组小鼠未见其它脏器肿瘤转移。结论来那度胺在体外对人宫颈癌SiHa细胞生长无影响,对小鼠SiHa细胞移植肿瘤具有生长抑制作用,可减少移植肿瘤组织中血管生成,并对肿瘤的转移起到一定抑制作用。Objective To study the effect of lenalidomide on human cervical cancer SiHa cells and mice transplanted tumor in vitro and in vivo,and to clarify the inhibitory effect and mechanism of lenalidomide on tumor.Methods CCK method in vitro to study the influence of lenalidomide and DMSO on SiHa cell growth.To establish a mouse cervical cancer SiHa transplantation model,the mice were divided into experimental group and control group of 6 each.The mice in the experimental group were given intraperitoneal injection of lenalidomide(50 mg/kg)daily and the control mice were intraperitoneally injected with 1% DMSO daily for a total of 21 days.After three weeks,the mice were sacrificed and the tumor tissues were sacrificed.The tumor volume,tumor weight,tumor histopathological changes,tumor angiogenesis and tumor metastasis in the two groups were compared.Results Lenalidomide had no effect on the growth of SiHa cells in vitro(P〈0.05).Lenalidomide could inhibit the growth of mice transplanted tumor.The tumor volume and weight of transplanted mice in the experimental group were lower than those in the control group(P〈0.05).The number of angiogenesis in the experimental group was lower than that in the control group(P〈0.05).While the control group mice had lung metastasis,the experimental group did not have any metastasis.Conclusion Lenalidomide has no effect on the growth of human cervical cancer SiHa cells in vitro.It can inhibit the growth of SiHa cell transplanted tumor in mice,reduce the angiogenesis in the transplanted tumor tissue and inhibit the metastasis of the tumor.
