摘  要:目的:研究脾气虚证和脾不统血证模型大鼠脾脏组织能量代谢变化及其差异,为科学解释中医脾脏理论内涵提供基础资料。方法:构建脾气虚证和脾不统血证大鼠模型,测定各组大鼠脾脏ATP含量、线粒体呼吸链复合物(ComplexⅠ、ComplexⅡ、ComplexⅢ、ComplexⅣ)活性、柠檬酸合酶(CS)活性和mtDNA拷贝数。结果:脾气虚证大鼠和脾不统血证大鼠脾脏ATP含量显著降低,呼吸链复合物活性、CS活性和mtDNA拷贝数较正常大鼠均发生显著改变,脾不统血证大鼠脾脏ATP含量、CS活性和mtDNA拷贝数与脾气虚证大鼠存在显著差异。结论:脾气虚证大鼠和脾不统血证大鼠脾脏组织线粒体能量代谢发生障碍,其柠檬酸循环和氧化磷酸化功能受到损伤,而脾不统血证和脾气虚证线粒体能量代谢障碍程度的不同是二者存在差异的重要原因。本研究认为,对中医脾虚证脾脏组织能量代谢功能损伤的研究能够积极的揭示脾虚证的科学本质。Objective : To study the changes of energy metabolism and its differences in spleen tissue of spleen Qi defi- ciency syndrome and spleen deficiency and failure of control blood syndrome groups and provide the basic data for scien- tifically explaining the connotation of spleen theory. Methods : The model of spleen Qi deficiency syndrome and spleen de- ficiency and failure of control blood syndrome were established. The levels of ATP, the activity of mitochondrial respirato- ry chain complex ( Complex I , Complex II, Complex III, Complex IV ) , the activity of citrate synthase (CS) and the mtDNA copy number were measured in the spleen of the rats. Results : The levels of ATP in the spleen of rats with spleen Qi deficiency syndrome and spleen deficiency and failure of control blood syndrome were significantly decreased. The ac- tivity of respiratory chain complex, CS activity and mtDNA copy number were significantly changed compared with those of normal rats. There were significant differences in ATP content, CS activity and mtDNA copy number between spleen deficiency and failure of control blood syndrome and spleen Qi deficiency syndrome rats. Conclu3ion: The energy metabo- lism of mitochondria in spleen tissue of rats with spleen Qi deficiency syndrome and spleen deficiency and failure of control blood syndrome were damaged, and the citric acid circulation and oxidative phosphorylation function are damaged. The difference of mitochondrial energy metabolism disorder between spleen deficiency and failure of control blood syn- drome and spleen Qi deficiency syndrome is an important reason for them. This study suggests that the study of energy metabolism in spleen tissue of spleen deficiency syndrome can positively reveal the scientific nature of spleen deficiency syndrome.
