摘 要:目的探讨乳腺癌肿瘤部位高表达趋化因子,并评估肿瘤部位高表达趋化因子与临床病理特征之问关系。体外验证趋化因子对乳腺癌细胞迁移运动的影响。方法收集乳腺癌患者标本72例。实时定量反转录聚合酶链反应(RT-qPCR)法检测趋化因子11(CCLll)/CXC型趋化因子配体(CXCL)8/CXCL9/CXCLIO/CXCLl2在乳腺癌组织和癌旁组织表达,筛选出肿瘤部位高表达趋化因子,并探讨其与临床病理特征间关系。体外Transwell技术检测趋化因子对乳腺癌细胞迁移运动能力的影响。结果趋化因子CXCLl0、CXCLl2在乳腺癌组织中高表达(1.18±2.16比0.54±1.52,t=2.328,P=0.023;1.33±1.46比0.78±1.03,t=2.408,P=0.019),但与患者年龄、肿瘤大小、TNM分期、淋巴结转移无明显相关性,不同人表皮生长因子受体2(Her-2)状态CXCLl0mRNA表达的差异有统计学意义(F=4.206,P=0.044),不同激素受体状态CXCLl2mRNA表达的差异有统计学意义(F=5.865,P=0.019)。两者均可提高乳腺癌细胞的迁移、侵袭能力。结论CXCLl0、CXCLl2在乳腺癌组织中高表达,影响患者疾病进展,并促进乳腺癌细胞迁移、侵袭。Objective To research highly expressed chemokines in breast cancer, evaluate the relationship between the chemokines with the clinicopathological factors, and validate the effects of chemokines on migration of breast cancer cells in vitro. Methods The specimens of 72 breast cancer patients were collected. The expression of chemokines (CCL) 11/chemokine ( C - X - C motif) ligand (CXCL) 8/ CXCL9/CXCL10/CXCL12) in breast cancer tissues and tissues adjacent to cancer was detected. The high- ly expressed chemokines were chosen in breast cancer and the relationship between the chemokines with the clinicopathological factors was evaluated. The effects of chemokines on the migration ability of breast cancer cells were analyzed by Transwell method in vitro. Results CXCL10 and CXCL12 were highly expressed in breast cancer ( 1.18 ±2. 16 vs. 0. 54 ± 1.52, t = 2. 328, P = 0. 023 ; 1.33 ± 1.46 vs. 0. 78 ± 1.03 ,t = 2. 408,P = 0. 019), which had no relevance to the patients' age, tumor size, TNM staging and the lymph node metastasis. The difference in CXCL10 mRNA expression in different states of human epidermal growth factor receptor - 2 ( Her - 2 ) and CXCL12 mRNA expression in various states of hormone receptors was statistically significant ( F = 4. 206, P = 0. 044 ; F = 5. 865, P = 0. 019 ). Both CXCL10 and CXCL12 could improve the migration and invasion ability of the breast cancer cells. Conclusion CXCL10 and CXCL12 were highly expressed in breast cancer, which would influence the progression of the disease and promote the migration and invasion of the breast cancer cells.
