摘 要:目的探寻凋亡抑制基因生存素(Survivin)在乳腺癌中的表达及其与细胞周期基因G1/S-特异性周期蛋白D1(Cyclin D1)、多药耐药基因p170表达的相关性。方法应用免疫组织化学方法,检测72例乳腺癌Survivin、Cyclin D1及p170基因的表达,另检测16例乳腺纤维腺瘤及12例癌旁正常组织中的Survivin基因表达,分析两两之间的相关性。结果乳腺癌Survivin、Cyclin D1及p170基因表达阳性率分别为84.7%(61/72)、88.9%(64/72)和93.1%(67/72)。Survivin与Cyclin D1及Survivin与p170表达之间均呈正相关(r=0.151,P=0.016;r=0.114,P=0.035);Cyclin D1与p170之间呈正相关(r=0.101,P=0.027),Survivin基因在乳腺纤维腺瘤中低表达18.75%(3/16),在乳腺癌旁正常组织中不表达0%(0/12)。结论Survivin、Cyclin D1及p170基因可能通过不同的途径发挥其肿瘤促进作用,三者在乳腺癌浸润、转移、复发及耐药方面参与乳腺癌的进展,针对上述三项基因的协同检测有助于乳腺癌预后的判断。Objective To investigate the expression of apoptosis inhibiting gene Survivin in breast cancer and the correlation of genes expression in cell cycle gene Cyclin D1 and multi - resistant gene plT0. Methods To apply immunohistochemical methods and detect the gene expression of Survivin, Cyclin DI and pl70 in 72 cases of breast cancer and detect the gene expression of the other 16 eases in breast fibroadenoma and of 12 cases in normal tissue adjacent to carcinoma, at the same time to analyze the correlation between the two. Results the gene expression positive rate of Survivin, Cyclin D1 and p170 expression in breast cancer was 84. 7% (61/72) , 88.9% (64/72) and 93.1% (67/72) respectively, there are positive correla- tions between Survivin and Cyclin D1 and between Survivin and p170 (r =0. 151, P =0. 016; r = 0. 114, P =0. 035) ; There is also a positive correlation relationship between Cyclin D1 and p170 (r = 0. 101, P = 0. 027) , there are a low expression of Snrvivin gene 18. 75% (3/16) in breast fibroadenoma and a zero ex- pression of Survivin gene 0. 00% (0/12) in normal tissue adjacent to breastcancer. Conclusion Survivin, Cyclin D1 and p170 genes may play a role of the tumor promotion through different ways, the three genes might participate in the progress of breast cancer in invasion, metastasis, recurrence and resistance, the col- laborative detection of the above three genes will contribute to breast cancer prognosis judgement.
