摘  要:目的检测微小RNA(miRNA,miR)-203mRNA在乳腺浸润性导管癌(IDC)及增生性病变中的表达,探讨在各病变中的作用和临床病理间的关系。方法应用一步法反转录一聚合酶链反应(RT-PCR)检测IDC及癌旁组织各60例及99例乳腺增生性病变和22例正常乳腺新鲜标本中miR-203mRNA表达;并结合临床病理特征分析。结果miR-203mRNA表达强度在正常乳腺组织、导管上皮普通性增生(UDH)、导管上皮非典型增生(ADH)和导管原位癌(DCIS)及IDC中分别为1.36、0.98、0.79、0.66和0.42,逐渐减弱且差异有统计学意义(P=0.005)。RT-PCR结果显示miR-203mRNA:(1)在IDC60例和癌旁组织中的表达水平分别为12%、89%,差异有统计学意义(P=0.003);(2)区域淋巴结转移:表达强度显示阳性组显著低于阴性组,NO〉N1〉N2期(P=0.000);(3)临床TNM分期:表达强度两两比较显示Ⅰ期〉Ⅲ期(P=0.000);(4)组织学分级中:表达强度Ⅰ级〉Ⅱ级〉Ⅲ级(P=0.000)。(5)雌激素受体(ER)、孕激素受体(PR)和人表皮生长因子受体2(C-erbB-2)表达的关系:表达强度在ER、PR、C-erbB-2(-)与ER、PR、C-erbB-2(++)、( )比较,差异有统计学意义(P=0.003)。结论在IDC和不同乳腺增生性组织中其蛋白表达强度存在差异。miR-203基因异常表达在IDC发生发展过程中起重要的调控作用。Objective To detect the microRNA (miRNA, miR) -203 mRNA expression in inva- sive ductal carcinoma of the breast ( IDC ) and adjacent non - cancerous lesions, and their relationships with clinical indicators of IDC and hyperplasia lesions were analyzed. Methods MiR - 203 mRNA of 60 breast IDC and 60 normal breast tissues and 99 breast hyperplasia lesions and 22 normal breast tissues were amplified by one -step reverse transcriptase -polymerase chain reaction (RT- PCR) method. Results RT - PCR showed miR - 203 mRNA : ( 1 ) The standardized coefficient were 1.36, 0. 98, 0. 79, 0. 66 and 0. 42, which were gradually lower in normal breast tissues, UDH, ADH, DCIS and IDC (P = 0. 005 ). The expression rate and standardized coefficient in IDC were significantly lower than those of breast tissues ( P = 0. 003 ). (2) The standardized coefficient in cases with axillary node metastasis was sig- nificantly lower than that in cases without axillary node matastasis, NO 〉 N1 〉 N2 (P = 0. 000). (3)The standardized coefficient at stage I was significantly higher than that at stage Ⅱ , and stage Ⅱ was signifi- cantly higher than stage Ⅱ ( P = 0. 000 ). ( 4 ) The standardized coefficient at grade I was significantly higher than that at grade Ⅱ , and grade 11 was significantly higher than grade m ( P = o. 000 ). ( 5 ) The standardized coefficient at estrogen receptor ( ER), progesterone receptor (PR) , C - erbB - 2 ( - ) was significantly higher than ER, PR, C - erbB - 2 ( ) , ( ) ( P = 0. 003 ). Conclusion The miR -203 protein expression in IDC and hyperplasia lesions have different levels, miR -203 may play a important role in IDC.
