摘 要:目的探讨大鼠局灶性脑缺血再灌注损伤白藜芦醇与热休克蛋白(HSP)70的关系和作用。方法随机设立假手术组、对照组和白藜芦醇预处理组3个组,后两组采用线栓法建立大鼠大脑中动脉闭塞模型(MCAO)缺血2 h再灌注,且设立6 h和24 h两个再灌注时间点。白藜芦醇预处理组按40 mg/kg经腹腔注射。分别在术后不同时间点进行神经功能学评分,采用免疫组化和免疫印迹法测定HSP70蛋白表达。结果对照组和白藜芦醇预处理组与假手术组比较神经功能评分显著增高,白藜芦醇预处理组又明显低于对照组(P<0.05);免疫组化和免疫印迹结果一致,对照组和白藜芦醇预处理组与假手术组比较HSP70蛋白表达显著增高,白藜芦醇预处理组HSP70蛋白表达又明显高于对照组(P<0.05)。结论大鼠局灶性脑缺血再灌注损伤白藜芦醇的神经保护作用可以通过热休克蛋白HSP70蛋白的表达增多来产生。Objective To investigate the relationship and use of resveratrol and heat shock protein(HSP)70 in focal cerebral ischemia reperfusion injury in rats. Methods SD rats were divided into randomly sham,control and resveratrol pretreatment 3 groups. Two groups were subdivided into two subgroups:6-hour and 24-hour reperfusion subgroup according to the reperfusion time after ischemia. Two groups except sham were established model of Middle cerebral artery occlusion(MCAO)by using suture embolus method,and reperfusion after ischemia 2 h. According to 40 mg/kg resveratrol pretreatment group intraperitoneal injection of resveratrol. The experimental models were observed for neurofunctional score at the different time points after operation,the expression of HSP70 protein were determined with immunohistochemical and westernblot method. Results Compared with the sham,the neurofunctional score was significantly higher about the control and resveratrol pretreatment groups,the resveratrol pretreatment group was significantly lower than the control group(P〈0.05).The results of immunohistochemical and western-blot method was accordance,the expression of HSP70 protein about the control and resveratrol pretreatment groups was significantly higher than the sham groups. The expression of HSP70 protein of the resveratrol pretreatment group was significantly enhanced than the control group(P〈0.05). Conclusion The neuroprotective effect of resveratrol were generated in focal cerebral ischemia reperfusion injury in rats,according to increasing the expression of HSP70 protein.
