摘 要:目的目前兔骨肿瘤模型的制作主要通过细胞悬液法或组织块植入法,但是哪种方法在制作肿瘤模型方面更为理想鲜见报道。本实验通过对比细胞悬液法及组织块植入法制作兔VX2骨肿瘤模型,旨在选择较为合理的造模方法。方法取传代VX2肿瘤荷瘤兔,将肿瘤制备成肿瘤组织块及细胞悬液,将瘤块植入20只新西兰兔左胫骨髓腔内,将肿瘤细胞悬液注入此20只家兔右胫骨内制备VX2骨肿瘤模型。于模型制备后第7、14和21天行双侧胫骨X射线检查,观察肿瘤生长速度和形态;模型制备后第21天处死动物,取肿瘤,观察其形态并取穿刺骨孔周围软组织行病理检查,明确有无肿瘤细胞转移。结果 2种方法建模成功率均为100%。建模后第7、14和21天X射线示瘤块植入组肿瘤生长迅速,形态较规整,而细胞悬液组肿瘤生长较慢。模型制备后21d,瘤块组发现穿刺孔周围软组织有肿瘤细胞浸润3只,而细胞悬液组有9只,瘤块植入组外渗率为15%,与细胞悬液植入组外渗率45%比较,差异有统计学意义,χ2=4.28,P<0.05。结论瘤块植入法较细胞悬液法制作骨肿瘤模型更为方便,肿瘤生长快,发生肿瘤细胞外渗概率小,是一种理想的骨肿瘤模型制备方式。OBJECTIVE The VX2 bone tumor model was established in rabbits with the cell suspension method or tumor tissue embedding method,but it was difficult to know which was better. This study compared the cell suspension method and tumor tissue embedding method in establishment of VX2 bone tumor model in rabbits. METHODS After separating the VX2 tumor from cancer bearing VX2 rabbit, the tumor tissue blocks and cell suspension were prepared;the tissue blocks was embedded into the left tibia(the issue block group) and the cell suspension was injected into the right tibia(the cell suspension group) of 20 rabbits. The X-Rays were used to observe the growth rate and the shape of the tumor at 7,14 and 21 d after model establishment. The rabbits were sacrificed and the tumors were taken out to observe the shape at 21 d. The soft tissue surrounding the puncture holes was taken to carry out the pathological examination. RESULTS The success rates were both 100% for two methods. The tumor in tissue block group was significantly greater and grew quickly than that in cell suspension group at 7,14 and 21 d after model establishment. Twenty-one days after model establishment, there were 3 cases that we could find the tumor cells in the soft tissue around the puncture hole in is- sue block group,but 9 cases in the cell suspension group. The tumor cells spreading of the issue block group and the cell suspension group were 15% and 45%, and the difference was statistically significant (χ^2= 4. 28, P 〈 0. 05). CONCLUSION The tissue embedding method is more convenient in operation and less tumor cells spreading in establishment of bone tumor model compared with cell suspension method.
