摘 要:以抗逆性较强的荒漠植物花花柴为材料,利用数字基因表达谱技术对用质量百分数20%的PEG溶液处理0、4、8、12和24 h后的花花柴幼苗的m RNA-Seq测序结果进行分析。结果表明,花花柴幼苗叶部在不同处理时间均上调表达的基因有122个,下调表达的基因54个,根部在不同处理时间均上调表达的基因73个,下调表达的基因79个。利用q RT-PCR和RT-PCR对可能与花花柴耐旱相关的6个差异表达基因进行了验证,发现其表达模式与表达谱结果一致,推测这些基因可能均与花花柴响应干旱胁迫相关。本研究为耐旱相关基因的发掘及应用提供了基础。In this study, the gene expression profile of Karelinia caspica seedling was used for analysis under treatment with 20% PEG at 0, 4, 8, 12, and 24 h through mRNA-Seq technology. The results indicated that there were 122 genes up-regulated and 54 genes down-regulated expression in the leaves, and 73 genes up-regulated and 79 genes down-regulated expression in the roots of Karelinia caspica seedling under different time treatments. And then, 6 genes with differential expression were tested by qRT-PCR and RT-PCR. The result showed that the expression pattern of these 6 genes were as same as the mRNA-Seq. It was inferred that these genes might be related with drought stress in Karelinia caspica. This research provides a basis for the exploration and application of drought related genes from Karelinia caspica.
