摘  要:目的 观察Caspase-1在大鼠缺氧相关性肺高压中的作用及茶多酚对Caspase-1的影响,探讨茶多酚防治缺氧相关性肺高压的可能机制.方法 将24只实验大鼠随机分为对照组(n=8)、肺高压组(n=8)和茶多酚组(n=8).肺高压组和茶多酚组建立肺高压模型,茶多酚组加用茶多酚200 mg·kg-1·d-1干预.4周后测量大鼠平均肺动脉压(mPAP),采用ELISA法检测血浆hs-CRP、IL-1β、Caspase-1浓度,RT-PCR检测肺组织Caspase-1 mRNA表达,Western blot检测肺组织Caspase-1蛋白表达.结果 肺高压组大鼠mPAP[(25.50±3.02)mm Hg]、血浆hs-CRP[(70.01±12.39)ng/L] 、IL-1β[(91.71 ±16.04)ng/L]、Caspase-1浓度[(134.06±16.03)ng/L]、肺组织Caspase-1 mRNA(1.239±0.304)和Caspase-1蛋白表达(0.329±0.051)均较对照组mPAP[(15.13± 1.96)mm Hg]、血浆hs-CRP[(20.68±4.29)ng/L]、IL-1β[(24.36±6.07)ng/L]、Caspase-1浓度[(31.64±5.41)ng/L]、肺组织Caspase-1 mRNA (0.708±0.138)和Caspase-1蛋白表达(0.118±0.015)明显升高(P均<0.01).茶多酚组mPAP[(20.50±0.93)mm Hg]、血浆hs-CRP[(49.16±9.02)ng/L]、IL-1β[(62.49± 17.03)ng/L]、Caspase-1浓度[(78.40±17.07)ng/L]、肺组织Caspase-1 mRNA(0.851±0.190)和Caspase-1蛋白表达(0.160±0.038)均较肺高压组明显降低(P均<0.01).相关分析显示,血浆Caspase-1与hs-CRP(r=0.784,P=0.000)和IL-1β(r=0.857,P=0.000)均呈正相关,肺组织Caspase-1蛋白表达量与mPAP呈正相关(r=0.809,P=0.000).结论 Caspase-1通过调控炎症介质功能参与大鼠缺氧相关性肺高压的形成.通过降低血浆Caspase-1浓度和下调肺组织Caspase-1表达,进而抑制肺血管炎症,可能是茶多酚改善缺氧相关性肺高压的作用机制之一.Objective To investigate the role of Caspase-1 in rats with pulmonary hypertension induced by hypoxia and the effects of tea polyphenols on Caspase-1.Methods 24 male Sprague Dawley rats were divided into 3 groups:control group(n=8),pulmonary hypertension group(n=8) and tea polyphenols group(n=8).Rats in the tea polyphenols group were treated with tea polyphenols (200 mg·kg-1·d-1).The rats were feed for 4 weeks.Then the mean pulmonary artery pressure(mPAP),the levels of hs-CRP,IL-1β and Caspase-1 in blood plasma,the expressions of Caspase-1 in pulmonary tissue were detected.Results In control group,the mPAP was (15.13±1.96)mm Hg;the levels of hs-CRP,IL-1βand Caspase-1 in blood plasma were (20.68±4.29)ng/L,(24.36±6.07)ng/L and (31.64±5.41)ng/L;the expressions of Caspase-1 mRNA and protein were (0.708±0.138)and (0.118±0.015).Compared with control group,mPAP (25.50±3.02)mm Hg,the levels of hs-CRP (70.01±12.39)ng/L,IL-1β(91.71±16.04)ng/L and Caspase-1 in blood plasma(134.06±16.03)ng/L,the expressions of Caspase-1 mRNA (1.239±0.304) and protein (0.329±0.051) were significantly increased in pulmonary hypertension group (all P〈0.01).The mPAP (20.50±0.93)mm Hg,the levels of hs-CRP (49.16±9.02)ng/L,IL-1β(62.49±17.03)ng/L and Caspase-1 (78.40±17.07)ng/L in blood plasma,the expressions of Caspase-1 mRNA (0.851±0.190) and protein (0.160±0.038) in tea polyphenols group were significantly decreased than those in pulmonary hypertension group(all P〈0.01).The level of Caspase-1 was positively correlated with the level of hs-CRP (r=0.784,P=0.000) and IL-1β(r=0.857,P=0.000),the expression of Caspase-1 protein was positively correlated with mPAP.Conclusion By promoting the inflammation of pulmonary artery,Caspase-1 involves in the process of pulmonary arterial hypertension.Tea polyphenols could regulate the level of Caspase-1 in blood plasma and the expressions of Caspase-1,which may be linked wit
