摘 要:目的观察微小RNA(miR)-27a-3p在肺腺癌组织中的表达以及对肺腺癌细胞增殖和迁移能力的影响。方法收集21例肺腺癌病人癌组织和癌旁组织,通过实时定量聚合酶链反应(RT-q PCR)技术检测miR-27a-3p的表达水平。采用细胞增殖实验(MTT法)和划痕实验观察miR-27a-3p过表达或干扰对肺腺癌细胞株A549增殖和迁移能力的影响。结果与癌旁组织比较,miR-27a-3p在肺腺癌组织中的表达水平显著上升(P<0.01)。细胞增殖及划痕实验结果显示,细胞转染48、72、96 h后,过表达miR-27a-3p后A549细胞的增殖、迁移能力显著高于对照组(P<0.05)。细胞增殖及划痕实验结果显示,细胞转染48、72、96 h后,沉默miR-27a-3p后A549细胞的增殖、迁移能力较对照组细胞显著降低(P<0.05)。结论 miR-27a-3p在肺腺癌中高表达,且其表达与肺腺癌细胞的增殖和迁移能力有关,提示miR-27a-3p可能参与调节肺腺癌细胞的生物学行为。Objective To investigate the expression of miR-27 a-3 p in lung adenocarcinoma tissue and the effect of miR-27 a-3 p on the proliferation and migration of lung adenocarcinoma cells. Methods Tumor tissue samples and adjacent tissue samples were collected from 21 patients with lung adenocarcinoma,and RT-q PCR was used to measure the expression of miR-27 a-3 p. MTT assay and wound healing assay were used to measure the effect of miR-27 a-3 p overexpression or interference on the proliferation and migration of lung adenocarcinoma A549 cells. Results Compared with the adjacent tissue samples,the lung adenocarcinoma tissue samples had a significant increase in the expression of miR-27 a-3 p( P〈0.01). The MTT assay and wound healing assay showed that at 48,72,and 96 hours after transfection,the miR-27 a-3 p overexpression group had significantly greater abilities of proliferation and migration than the control group( P〈0.05),and the miR-27 a-3 p silencing group had significant reductions in proliferation and migration than the control group( P〈0.05). Conclusion MiR-27 a-3 p is highly expressed in lung adenocarcinoma,and its expression is associated with the proliferation and migration of lung adenocarcinoma cells,suggesting that miR-27 a-3 p may be involved in the regulation of the biological behavior of lung adenocarcinoma cells.
