摘 要:采用不同浓度的NaCl,运用正交试验法对黑大豆种子的球蛋白进行提取,并进行了SDS-PAGE实验。结果显示:对提取影响最大的因素在不同提取体系中均为固液比,而影响最小的因素在不同的体系有所不同,其中两组为提取液浓度,一组提取体系为温度。黑大豆种子球蛋白电泳最优化的体系是NaCl浓度为4.0%、固液比为1:7、提取温度为50℃、提取时间为2 h、上样量为10μL的电泳体系。Using NaCl of different concentrations,the globulin of black soybean seeds was extracted by orthogonal test,and the SDS-PAGE experiment was carried out.Results showed: For different extracting systems,the solid-to-liquid Ratio was the maximum influencing factor, but minimal impact factors vary in different system,including two groups for the extract concentration. one was the temperature for a set of extraction system. Black soybean seed globulins electrophoresis optimization system was4.0% NaCl concentration,solid-to-liquid ratio was 1: 7,extraction temperature was 50 ℃,extraction time was 2 h,the loading sample amount was 10μL electrophoresis system.
