COXⅣ的表达与结直肠癌患者预后的相关性

摘　　要：目的检测COXⅣ基因在结直肠癌(colorectal cancer,CRC)患者组织及细胞中的表达水平,分析其与预后的相关性。方法用实时定量PCR和Western blot的方法检测COXⅣ在23对肠癌组织及癌旁组织,以及6个CRC细胞系中的表达,运用统计学Pearson卡方检验分析组织及细胞表达的相关性。结果在23对CRC组织中COXⅣm RNA和蛋白水平的表达同癌旁相比显著升高(P=0.05)。在结直肠癌细胞系中,COXⅣ的m RNA表达水平明显升高,呈阳性表达。COXⅣ在CRC癌组织中的表达相对高于对应癌旁组织(P=0.033),结直肠癌患者COXⅣ表达与预后之间无显著的相关性。结论 COXⅣ在CRC患者癌组织中表达明显升高,COXⅣ过表达与CRC患者预后无显著相关性。Objective To explore the correlation between COX Ⅳ gene expression and prognosis in patients with colorectal cancer（ CRC）. Methods By real-time quantitative PCR and Western blot method,COX Ⅳ was detected in 23 pairs of cancer tissues and para-carcinoma tissues and 6 CRC cell lines. Results The m RNA and protein expression levels of COX Ⅳ were significantly higher in CRC tissues than in para-carcinoma tissues（ P = 0. 05）. In colorectal cancer cell lines,the COX Ⅳ m RNA expression was positive and increased significantly. The expression of COX Ⅳ was higher in CRC cancer tissues than that in corresponding para-carcinoma tissues（ P = 0. 033）. There was no significant correlation between COX Ⅳ expression and prognosis of colorectal cancer patients. Conclusion COX Ⅳ has been significantly elevated in CRC patients,but the overexpression of COX Ⅳ is not associated with prognosis in patients with CRC.
