摘  要:目的:探讨游泳运动对小鼠急性肺损伤的影响,分析运动改善急性肺损伤的可能作用机制。方法:构建LPS诱导的急性肺损伤模型,通过H&E染色、肺湿干比、MPO活性、荧光定量PCR及免疫印迹等方法判断炎症的严重程度。结果:1)H&E染色结果显示,运动干预后,LPS诱导的炎症症状减轻。2)运动干预后,肺湿干比具有显著性差异(P<0.05),肺灌洗液(BALF)总蛋白浓度显著减低(P<0.01),MPO活性显著降低(P<0.001)。3)LPS运动训练组CD11b+Gr-1+细胞比率明显低于LPS对照组。4)RT-PCR检测促炎因子(包括IL-1β、TNF-α和IL-6),结果显示LPS运动训练组促炎因子m RNA水平低于LPS对照组,且具有显著性差异(P<0.05)。5)运动训练后,显著降低P38 MAPK磷酸化水平(P<0.01)。结论:游泳运动能降低LPS诱导的炎性细胞浸润和促炎因子的表达,同时可以减轻肺组织通透性及肺组织损伤从而改善LPS引起的急性肺损伤。Objective: The purpose of the study was to determine the role of swimming in acute lung injury, and explore the possible mechanisms. Methods: To build the model of acute lung injury induced by LPS, the severity of inflammation was determined by HE staining, lung wet dry ratio, MPO activity,fluorescence quantitative PCR and immunoblotting. Results: 1) The results of HE staining showed that LPS induced inflammatory symptoms were reduced after exercise. 2) After exercise, there was a significant difference in lung wet dry ratio(P〈0.05), total protein concentration in lung lavage fluid(BALF) was significantly decreased(P〈0.01), and MPO activity was significantly decreased(P〈0.001).3) The ratio of CD11 b+Gr-1+ cells in the LPS exercise group was significantly lower than that of the LPS control group 4)Compared to groups A and B, the m RNA expression of IL-1β,TNF-α and IL-6 in group C were increased significantly, while the IL-1β,TNF-α and IL-6 m RNA expression in group D was decreased significantly(P〈0.05). 5) After exercise, the phosphorylation level of P38 MAPK was significantly reduced(P〈0.01).Conclusions: 6-week regular swimming exercise can relieve LPS-induced ALI by inhibit the infiltration of inflammatory cells, the expression of pro-inflammatory cytokines, the permeability and injury of lung tissue.
