摘 要:目的探讨生长分化因子11(growth differentiation factor 11,GDF11)在胰腺癌中的表达及意义。方法收集解放军总医院肝胆外二科2016年12月-2017年3月胰腺切除手术的28例胰腺癌组织及对应癌旁组织,以及购于上海芯超公司组织芯片中的63例胰腺癌及对应癌旁组织。通过实时定量PCR检测28对胰腺癌和对应癌旁组织中的m RNA表达量,免疫组化法检测91对(包括自取的28对与组织芯片中的63对)胰腺癌及对应癌旁组织中的GDF11蛋白的表达情况,并分析GDF11的蛋白表达水平与临床病理指标之间的关系。结果 GDF11的m RNA水平在癌旁组织中的表达量显著高于癌组织中的表达(P=0.003);免疫组化结果显示胰腺癌组织中GDF11蛋白的高表达率为40.7%,显著性低于癌旁组织中的高表达率(71.4%)(P<0.001);GDF11的蛋白表达水平与年龄、性别、组织学分级和远处转移与否不相关(P均>0.05),GDF11的高表达与神经浸润较少(P=0.004)、T分期较低(P=0.003)、N分期较低(P=0.018)相关。结论 GDF11在胰腺癌中表达量下调,且GDF11的表达升高可能抑制胰腺癌的进展。Objective To investigate the expression and significance of growth differentiation factor 11 (GDF11) in pancreatic cancer. Methods Pancreatic cancer and corresponding paracancerous tissues in 28 patients who underwent pancreatectomy in the No.2 department of hepatobiliary and pancreatic surgical oncology in Chinese PLA General Hospital from December 2016 to March 2017, and another 63 pairs of pancreatic cancer and corresponding paracancerous tissues from tissue microarray of Shanghai Outdo Biotech Company were collected. The mRNA expression levels in 28 pairs of pancreatic cancer and corresponding paracancerous tissues were detected by real-time quantitative PCR, and the expression of GDF11 protein in 91 pairs (including 28 pairs of self-picking and 63 pairs in the tissue microarray) of pancreatic cancer and corresponding paracancerous tissues was detected by immunohistochemistry. Moreover, the relationship between GDF11 protein expression and clinicopathological parameters was analyzed. Results The mRNA level of GDF11 was significantly higher in the paracancerous tissues than in the cancer tissues (P=0.002 5). The results of immunohistochemistry showed that the high expression rate of GDF11 protein in pancreatic cancer tissues was significantly lower than that in adjacent normal tissues (40.7% vs 71.4%, P 〈 0.001). The protein expression levels of GDFll showed no significant difference between two groups in age, gender, histological staging and metastasis, while the higher expressions of GDF11 were associated with less perineural invasion (P=0.004), lower T stage (P=0.003) and lower N stage (P=0.018). Conclusion The expression of GDF11 is down-regulated in pancreatic cancer and elevated expression of GDF11 may suppress the development of pancreatic cancer.
