甘草附子汤对佐剂性关节炎小鼠的治疗作用

摘　　要：目的：研究甘草附子汤对佐剂性关节炎（AA）小鼠的作用研究。方法：雄性Balb/c小鼠随机分为6组,为正常组、模型组、雷公藤多苷组、甘草附子汤高、中、低剂量组（3.9,7.8,15.6 g·kg^-1）,每组12只。除正常组外,采用AA小鼠模型,造模第12天,灌胃给予相应药物,1次/d,连续18 d,观察药物对AA小鼠足肿胀的影响;苏木素-伊红（HE）染色观察踝关节组织病理学改变;通过酶联免疫吸附法（ELISA）检测AA小鼠血清中的细胞因子白细胞介素-1β（IL-1β）,白细胞介素-6（IL-6）,肿瘤坏死因子-α（TNF-α）的水平;蛋白免疫印迹法（Western blot）检测滑膜组织基质金属蛋白酶-1（MMP-1）和基质金属蛋白酶-3（MMP-3）蛋白表达。结果：与模型组比较,甘草附子汤可抑制AA小鼠足肿胀,改变炎细胞浸润,减轻滑膜增生和骨侵蚀,减少AA小鼠血清中炎症因子TNF-α,IL-1β和IL-6水平（P〈0.05）,降低AA小鼠滑膜中MMP-1和MMP-3蛋白表达（P〈0.05）。结论：甘草附子汤AA小鼠具有明显治疗作用,其机制可能与降低炎症因子的产生和降低滑膜组织的MMP-1及MMP-3表达有关。Objective： To investigate the effects of Gancao Fuzi Tang on adjuvant-induced arthritis（ AA）in mice. Method： Male Balb/c mice were randomly divided into 6 groups： normal group,model group,glycoside from Tripterygium wilfordii group,Gancao Fuzi fang high dose,middle dose and low dose groups（ 3. 9,7. 8,15. 6 g·kg^-1）,n = 12 in each group. AA model was used for all the mice except those in normal group,and on day 12 after modeling,corresponding drugs were given,qd,for 18 days. Then the effect of drugs on paw edema of the animals was observed and the histopathological changes of the ankle joint were observed by hematoxylin-eosin（ HE） staining. The levels of interleukin-6（ IL-6）,interleukin-1β（ IL-1β） and tumor necrosis factor-α（ TNF-α）in serum were examined by enzyme-linked immunosorbent assay（ ELISA）. The protein expression levels of matrix metalloproteinases-1（ MMP-1） and matrix metalloproteinases-3（ MMP-3） were detected by Western blot.Result： Gancao Fuzi Tang significantly alleviated joint swelling,changed inflammatory cell infiltration,alleviated synovial hyperplasia and bone erosion,reduced the levels of TNF-α,IL-1β and IL-6（ P〈0. 05）,and decreased MMP-1 and MMP-3 protein expressions（ P〈0. 05） in AA mice. Conclusion： The results indicate that Gancao Fuzi Tang has therapeutic effect on AA mouse,and the mechanism might be associated with its anti-inflammatory effects and down-regulating MMP-1 and MMP-3 expressions in synovial tissues.
