摘  要:目的观察三七总皂苷(PNS)是否可通过抑制低氧诱导因子(HIF-1α)减轻单侧输尿管梗阻(UUO)大鼠肾间质纤维化(RIF)。方法 SD大鼠随机分为假手术组、UUO组、PNS组,每组10只;造模成功后PNS组给予PNS100 mg/(kg·d)腹腔注射,14 d后处死3组大鼠,留取大鼠血标本检测血尿素氮、血肌酐、胱抑素C;用ELISA法检测血清HIF-1α水平;UUO组织行HE染色、Masson染色,通过肾小管间质损害程度评分、RIF半定量分析积分值观察肾间质损害程度。结结果与假手术组比较,UUO组血尿素氮、血肌酐、胱抑素C、HIF-1α均明显升高,差异有统计学意义(P<0.05);与UUO组比较,PNS组治疗后血尿素氮、血肌酐、胱抑素C、HIF-1α均下降,差异有统计学意义(P<0.05);肾脏组织HE、Masson染色均显示:与假手术组比较,UUO组肾小管间质损害程度明显增加(P<0.05);与UUO组比较,PNS组肾小管间质损害程度明显减轻(P<0.05)。结论三七总皂苷可改善UUO大鼠RIF程度,并降低HIF-1α水平,可能是通过抑制HIF-1α减轻RIF。Objective To observe the effect of panax notoginseng saponins(PNS) reducing the renal interstitial fibrosis of unilateral ureteral obstruction(UUO) rats by inhibiting hypoxia-inducible factor-1α(HIF-1α).Methods SD rats were randomly divided into 3 groups: sham operation group, UUO group and PNS group.After successful modeling the PNS group was given PNS 100 mg/(kg · d) via intraperitoneal injection after UUO operation. All the rats were sacrificed on postoperative day 14 and specimens were collected. The blood content of urea nitrogen, serum creatinine and cystatin C were detected. The serum level of HIF-1α was tested by ELISA. The level of renal interstitial fibrosis was observed under a light microscope to evaluate it with the hematoxylin eosin(HE) staining and Masson staining. Results Compared with the sham operation group,the levels of urea nitrogen, serum creatinine and cystatin C of the UUO group were significantly increased(P〈0.05), the difference was statistically significant. And the Masson staining renal tissue was displayed.Compared with the sham operation group, the renal interstital fibrosis degree was significant increased(P〈0.05), but compared with the UUO group, the renal interstital fibrosis degree was markedly decreased in the PNS group(P〈0.05). Conclusion PNS can improve the renal interstitial fibrosis of UUO rats and inhibiting HIF-1α maybe a possible mechanism.
