摘  要:目的探讨miRNA(miR)-429基因表达及其启动子甲基化在乳癌中的作用。方法收集50例乳腺浸润性癌病人的手术切除标本,采用实时荧光定量PCR检测miR-429基因在癌组织和癌旁组织中的表达情况,采用甲基化特异性PCR检测癌组织和癌旁组织中miR-429基因启动子区CpG岛的甲基化水平,并分析miR-429基因启动子区甲基化状态与乳癌临床病理特征的关系。结果与癌旁组织相比,癌组织中miR-429基因的表达水平明显下调(t=19.227,P<0.01),miR-429基因启动子区CpG岛的甲基化水平明显升高(χ~2=5.392,P<0.05)。miR-429基因启动子区甲基化水平与乳癌肿瘤大小及淋巴结转移相关(χ~2=5.236、6.202,P<0.05)。结论 miR-429基因异常高甲基化可能导致miR-429基因低表达,参与乳癌的发生发展。Objective To explore the expression of miRNA(miR)-429 and the role of its promoter methylation in breast cancer. Methods The surgical specimens from 50 patients with invasive breast cancer were collected.The expression of miR-429 in cancerous and para-cancerous tissues was measured by quantitative real-time PCR,and the methylation status of the CpG islands around the promoter region of miR-429 was investigated by methylation-specific PCR.The relationship between the methylation status of the promoter region of miR-429 and the clinicopathological features of breast cancer was analyzed. Results Compared with the para-cancerous tissues,the cancerous tissues had significantly reduced expression of miR-429(t=19.227,P〈0.01),as well as significantly increased methylation of the CpG islands around the promoter region of miR-429(χ^2=5.392,P〈0.05).The methylation status of the promoter region of miR-429 was correlated with primary tumor size and lymph node metastasis(χ^2=5.236 and 6.202,P〈0.05). Conclusion The increased methylation of the promoter region of miR-429 may lead to reduced miR-429 expression,which is involved in the tumorigenesis and progression of breast cancer.
