摘 要:目的构建人端粒酶逆转录酶(hTERT)启动子调控的肿瘤坏死因子相关凋亡诱导配体(TRAIL)基因表达载体,探讨其对结肠癌细胞HT-29增殖的抑制作用。方法构建重组质粒pshuttle-TRAIL和pshuttle-hTERT-TRAIL,通过脂质体转染结肠癌HT-29细胞,采用MTT法检测重组质粒对细胞增殖的影响及流式细胞术检测细胞凋亡的变化。结果 pshuttle-TRAIL和pshuttle-hTERT-TRAIL组细胞增殖率低于空白对照组和空质粒转染组;流式细胞术检测转染后细胞凋亡率升高,并且pshuttlehTERT-TRAIL凋亡率高于pshuttle-TRAIL组。结论 hTERT启动子调控的TRAIL可明显抑制结肠癌细胞增殖以及诱导细胞凋亡,可能是结肠癌靶向治疗的新途径。Objective To construct the hTERT promoter-driven TRAIL expression vector and explore its inhibitory effects on the proliferation of colonic cancer cells. Methods The recombination of pshuttle-Trail and pshuttle-hTERT-TRAIL were constructed and transfected into HT-29 cells through liposome. MTT method was used to detect the effects of the recombinat plasmids on cell proliferation in transfected cells. Flow cytometry was used to detect the changes of apoptosis. Results The anti-proliferative effect in the pshuttle-TRAIL and pshuttle-hTERT-TRAIL groups were significantly higher than in the blank control group and empty-plasmid transfection group. Flow cytometry showed higher apoptotic rate in the pshuttle-hTERT-TRAIL group than in the pshuttle-TRAIL group. Conclusion The TRAIL regulated by the hTERT promoter can significantly inhibit the proliferation of colon cancer cells and induce apoptosis, which may be a new targeted therapy for colon cancer
