摘  要:目的探讨血脂康对非酒精性脂肪性肝病大鼠肝炎症及氧化应激的影响,并分析其相关机制。方法将32只Wistar大鼠分为对照组、模型组、洛伐他汀组、血脂康组,每组各8只。对照组给予普通饲料喂养,其他3组给予高脂饲料喂养,高脂饲料喂养第1天,一次性给予维生素D3(6×10~5U/kg)腹腔注射,对照组给予同等剂量的生理盐水腹腔注射;喂养6周后,洛伐他汀组、血脂康组分别给予洛伐他汀2.5 mg/(kg·d)、血脂康300 mg/(kg·d)灌胃治疗。喂养12周后,检测各组大鼠的血脂、肝功能;ELISA法检测大鼠血清肿瘤坏死因子(TNF)-α和白细胞介素(IL)-6水平;免疫组织化学方法检测肝组织转化生长因子(TGF)-β的表达;观察肝组织的形态学变化。结果模型组大鼠的甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、TNF-α、IL-6、谷丙转氨酶(ALT)、谷草转氨酶(AST)水平均高于洛伐他汀组和血脂康组;高密度脂蛋白胆固醇(HDL-C)水平低于洛伐他汀组和血脂康组。洛伐他汀组TNF-α、IL-6水平均高于血脂康组,两组间比较,差异有统计学意义(P<0.05)。血脂康组和洛伐他汀组在血脂水平、肝功能、肝组织的病理学改变等方面比较,差异均无统计学意义(P>0.05)。模型组大鼠的肝脂肪变性和胶原增生最严重,对照组最轻。模型组大鼠肝组织的TGF-β阳性表达最多,对照组最少。结论血脂康能够调节血脂,具有抗炎作用,并能通过抗炎机制对肝组织发挥保护作用,其效果优于洛伐他汀。Objective To investigate the effect of Xuezhikang on hepatitis and oxidative stress in rats with nonalcoholic fatty liver disease,and analyze its related mechanism. Methods A total of 32 Wistar rats were assigned randomly into 4 groups: the control group,model group,lovastatin group and Xuezhikang group,with 8 rats in each group. Basic chow was provided for control group,the rats in the other three groups were fed with a high fat diet. On the first day,the rats were injected with a single dose of vitamin D3( 6 × 10~5 U/kg),the rats in the control group were injected with the same dose of saline. After 6 weeks of high-fat diet,the rats in the lovastatin group were treated with lovastatin 2. 5 mg/( kg·d) and the rats in the Xuezhikang group were treated with Xuezhikang 300 mg/( kg·d) by gastric tube. After 12 weeks of high-fat diet,the blood lipid and liver function of rats in each group were detected. Serum TNF-α and IL-6 levels were detected by ELISA; immunohistochemistry was used to detect the expression of TGF-β in liver tissues; morphological changes of liver tissue were observed. Results The TG,TC,LDL-C,TNF-α,IL-6,ALT and AST levels of the model group were higher than those of the lovastatin group and the Xuezhikang group; HDL-C level was lower than that of the lovastatin group and the Xuezhikang group. TNF-α and IL-6 levels in the lovastatin group were higher than those in the Xuezhikang group,and the difference between the two groups was statistically significant( P〈 0. 05). Comparison of serum lipid level,liver function and pathological changes of liver tissue between Xuezhikang group and lovastatin group had no statistically significant difference( P〉 0. 05). The hepatic steatosis and collagen hyperplasia were the most serious in the model group and the least in the control group. In the model group,the positive expression of TGF-β was the highest in the liver tissue of rats,while that of the control group was the least. Conclusion Xuezhikang can regulate blood lipids,h
