益气润目汤对干眼小鼠眼表组织IL-1β及P-JNK表达的影响

摘　　要：目的观察益气润目汤对高渗盐水诱导的干眼模型小鼠眼表组织中促炎因子白细胞介素-1β(IL-1β)、磷酸化氨基末端蛋白激酶(P-JNK)的影响。方法将雌性BALB/c小鼠随机分为6组:空白对照组、模型组、中药高剂量组、中药中剂量组、中药低剂量组、西药组,每组10只,除空白对照组,剩余小鼠均采用高渗盐水点眼诱导干眼动物模型,中药高剂量组、中药中剂量组、中药低剂量组分别灌胃给予含生药0.754 g·ml^(-1)、0.377 g·ml^(-1)、0.188g·ml^(-1)的中药益气润目汤,西药组给予玻璃酸钠滴眼液持续点眼,治疗7 d、14 d后检测每组角结膜组织中IL-1β、P-JNK的变化。结果免疫组化结果显示,中药各剂量组治疗7 d、14 d后,角结膜上皮IL-1β、P-JNK阳性表达(免疫组化)明显低于模型组(P<0.05);反转录聚合酶链反应(RT-PCR)结果显示,各中药组角结膜中IL-1β mRNA的表达水平亦较同期模型组明显减少(P<0.05)。结论益气润目汤能明显减少炎症相关因子的分泌,抑制炎症,减少眼表损伤,稳定泪膜。OBJECTIVE To observe the effects of Yi Qi Run Mu decoction on the expression of IL-1β、P-JNK in mice corneal epithelial cell response to dry eye model induced by hypertonic saline in the female mice. METHODS the female BALB/c mice were randomly divided into 6 groups： the blank control group,model group, high dose of Chinese traditional medicine group,middle dose of Chinese medicine group, low dose of Chinese traditional medicine group, the west-ern medicine group. hyaluronate sodium eye drops group. each group of 10, except the blank con-trol group, others adopt to be dry eye induced by hypertonic saline. High dose of Chinese tradition-al medicine group,middle dose of Chinese medicine group and low dose of Chinese traditional medicine group received intragastric infusion of Yi Qi Run Mu Decoction including 0.754 g·ml^-1、0.377 g·ml^-1、0.188 g·ml^-1 of Chinese traditional medicine,the western medicine group received hyaluronate sodium eye drops. Immunohistochemistry and PCR were performed to measure IL-1β and P-JNK in mice conjunctivitis epithelium after 7 and 14 days after treatment. RESULTS Immunohistochemical results showed that after treatment of 7 d and 14 d in each dose group of tradi-tional Chinese medicine, the positive expression of il-1 and p-jnk in the conjunctivitis epithelium （immunohistochemistry） was significantly lower than that in the model group（P〈0.05）. Reverse transcription polymerase chain reaction（RT-PCR）showed that the expression level of il-1 in the conjunctiva of Chinese medicine group was significantly lower than that in the model group（P〈0.05）.CONCLUSIONS Yi Qi Run Mu decoction can significantly reduce the secretion of inflammation-related factors, restrain inflammation, reduce the surface damage, and stabilize the tear film.
